Anhang","In der vorliegenden Arbeit wurde die Induktion der Apoptose durch die pro- apoptotischen Bcl-2-Proteine Bax und Nbk/Bik in Melanomzellen untersucht. Die Überprüfung der basalen Expressionsrate zeigte eine durchgehende Expression des pro-apoptotische Multidomänenproteins Bax in Melanomzelllinien, während eine Basisexpression des pro-apoptotische BH3-only-Proteins Nbk/Bik nur in einigen Zelllinien vorlag. Zur Überexpression von Bax und von Nbk in Melanomzellen wurde ein Tetracyclin/Doxycyclin-abhängiges Expressionssystems verwendet. Erstmalig zeigte sich hier, dass die Doxycyclin-abhängige Proteinexpression durch die Zugabe des Lösungsmittels DMSO (Dimethylsulfoxid) deutlich gesteigert werden konnte. Die kontrollierte Expression von Bax und von Nbk induzierte in verschiedenen Melanomzelllinien Apoptose, sowohl nach transienter als auch nach stabiler Transfektion. Durch die Expression von Bax und von Nbk wurden die Melanomzellen für eine zusätzliche Behandlung mit Chemotherapeutika bzw. mit einem agonistischen CD95-Antikörper signifikant sensitiviert. Insbesondere ließ sich in einem Mausmodell durch die induzierte Expression von Bax und von Nbk das Melanomwachstum in vivo wesentlich verzögern. Diese Befunde belegen, dass Proteine aus beiden Klassen der pro- apoptotischen Bcl-2-Proteine über das Potential verfügen, die Apoptoseresistenz von Melanomzellen zu überwinden und die Zellen für pro- apoptotische Behandlungen zu sensitivieren. Interessanterweise erwies sich die durch Nbk induzierte Apoptose in Melanomzellen, trotz eindeutiger Befunde hinsichtlich DNA-Fragmentierung und Chromatinkondensation, als weitgehend unabhängig von einer Caspasenaktivierung und der Freisetzung von Cytochrom C. Es fanden sich weder eine Prozessierung Apoptose-relevanter Caspasen noch konnten selektive Caspaseinhibitoren den pro-apoptotischen Effekt von Nbk verhindern. Im Gegensatz dazu zeigte sich nach Expression des CD95-Liganden (CD95L), der den Todesrezeptor-kontrollierten Apoptoseweg in Melanomzellen einleitet, eine deutliche Aktivierung von Caspase-3 und Caspase-8 sowie eine Sensitivität gegenüber der Inhibition von Caspasen. Die fehlende Freisetzung von Cytochrom C aus den Mitochondrien wies auf die mögliche Bedeutung anderer Zellorganellen in der durch Nbk induzierten Apoptose hin. Erstmals wurde in der vorliegenden Arbeit eine Relevanz der Lysosomen an apoptotischen Prozessen in Melanomzellen belegt. In den Lysosomen sind Cysteinproteasen der Cathepsinfamilie lokalisiert, die eine hohe Ähnlichkeit mit Caspasen aufweisen und unter physiologischen Bedingungen bei der intrazellulären Proteindegradation mitwirken. Durch Anfärbung der Lysosomen mit dem pH- indikator Acridin-Orange konnte eine Erhöhung ihres pH-Wertes nach Expression von Nbk nachgewiesen werden. Auch durch Expression von CD95L kam es zu diesem Anstieg, nicht jedoch nach Behandlung der Melanomzellen mit dem Chemotherapeutikum Doxorubicin. Weiterhin zeigte sich eine Freisetzung der Cathepsine B, L und H aus den Lysosomen in das Zytosol sowohl nach Expression von Nbk als auch von CD95L. In klarer Übereinstimmung mit einer Funktion der Cathepsine in der Apoptose führte die Inhibition von Cathepsin B und Cathepsin L zur Hemmung der durch Nbk oder CD95L induzierten Apoptose. Diese Daten belegen eine Bedeutung der lysosomalen Proteasen der Cathepsinfamilie sowohl in der Caspasen-unabhängigen als auch in der Caspasen-abhängigen Apoptose in Melanomzellen. Pro-apoptoische Bcl-2-Proteine ermöglichen durch ihre übergeordnete Rolle in den über Organellen kontrollierten Apoptosewegen eine gezielte Induktion sowohl Caspasen-abhängiger als auch Caspasen-unabhängiger Signalkaskaden. Sie verfügen damit über das Potential auch in Tumorzellen effizient Apoptose auslösen zu können.","Melanoma is a highly malignant tumor of the skin, characterized by a strong resistance to common therapeutical approaches. Its weak responsiveness to chemotherapy and immunotherapy is based on the pronounced apoptosis deficiency of melanoma cells. Apoptosis (programmed cell death) represents a general genetic safeguard mechanism of multicellular organisms for the elimination of superfluous or dangerous cells (virus-infected or cancer cells). Mitochondria are crucial regulators of apoptosis in several pathways and their function in apoptosis is tightly controlled by the family of Bcl-2 proteins. This large family contains both antiapoptotic and proapoptotic factors. Depending on the number of their BH domains (Bcl-2 homology), proapoptotic Bcl-2 proteins are further subdivided into multidomain proteins and BH3-only proteins. In the present study, the inducibility of apoptosis in melanoma cells by proapoptotic Bcl-2 proteins from both subgroups was investigated. The proapoptotic multidomain protein Bax revealed moderate basal expression levels in all melanoma cell lines, analysed, whereas the BH3-only protein Nbk/Bik was expressed only weakly in a few melanoma cell lines. Overexpression of Bax and of Nbk in melanoma cells was achieved by applying a tetracycline/doxycycline- regulateable expression system. Interestingly, application of the solvent DMSO (dimethylsulfoxide) was able to increase the doxycycline-dependent protein expression, as was shown here for the first time. The conditional expression of Bax and of Nbk strongly induced apoptosis in melanoma cells, after transient and after stable transfection. It also significantly sensitized melanoma cells for additional proapoptotic treatments as with chemotherapeutics or with agonistic CD95 antibody. In an in vivo nude mouse model, melanoma growth was strongly delayed by the expression of Bax or of Nbk. These findings clearly demonstrated the potential of proapoptotic Bcl-2 proteins to overcome apoptosis resistance of melanoma and to sensitize cells for therapeutic approaches. Nbk-induced apoptosis revealed broad hallmarks of apoptosis as DNA-fragmentation and condensation of nuclear chromatin. Interestingly however, apoptosis induction by Nbk was largely independent of caspase activation and of mitochondrial release of cytochrome c. Melanoma cells revealed neither processing of caspase-3 or caspase-8 nor sensitivity towards selective caspase inhibition, after Nbk expression. In clear contrast, caspase-3 and caspase-8 were activated in melanoma cells after induction of death receptor-controlled apoptosis by the expression of CD95 ligand (CD95L). Additionally, CD95L-induced apoptosis was also repressed by caspase inhibition. Analyses of the involvement of cell organelles in Nbk-induced apoptosis revealed for the first time an impact of lysosomes on melanoma cell apoptosis. In lysosomes, cysteine proteases of the cathepsin family are enclosed, which mediate protein degradation under physiological conditions. Staining of lysosomes with the pH indicator acridine orange revealed a significant increase of lysosomal pH after the expression of Nbk and of CD95L. On the other hand, treatment of melanoma cells with the chemotherapeutic drug doxorubicin showed no effect on lysosomal pH. Most significant, a release of cathepsins (B, L and H) into the cytosol was observed after the induction of Nbk as well as of CD95L. Further supporting the pivotal role of cathepsins in melanoma cell apoptosis, inhibition of cathepsin B and cathepsin L completely blocked apoptosis triggered by Nbk or by CD95L. These findings strongly suggest a decisive role of lysosomal proteases of the cathepsin family in caspase-independent as well as in caspase-enclosing apoptosis pathways in melanoma cells. Proapoptotic proteins of the Bcl-2 family exert important functions in organelle-controlled apoptosis. Thus, control of apoptosis by Bcl-2 proteins may also evolve as a powerful tool for overcoming therapy resistance of tumor cells, as shown here for melanoma cells.
