Welche Rolle dabei Hormone, wie Stress- bzw. gastrointestinale Peptidhormone, spielen, wird zurzeit aktiv erforscht und ist auch Gegenstand der vorliegenden Arbeit, wobei das Chemerin eine zentrale Rolle während dieser Studie einnimmt. Das Plasmaprotein Chemerin sowie sein Derivat mit Peptidstruktur Chemerin-9 besitzen unterschiedliche Funktionen im menschlichen Organismus. Als ein Chemokin ist Chemerin unter anderem in den Inflammationsprozess involviert und induziert die Adipo- und Angiogenese. Der Beitrag von Chemerin zur kolorektalen Kanzerogenese ist bisher nicht charakterisiert worden. In einem Screeningverfahren wurde die proliferative Wirkung von Stresshormonen Dexamethason und Noradrenalin sowie Peptiden Neurotensin, GLP-2, Somatostatin-Analogon SOM230 und Chemerin-9 untersucht. Dazu wurde DAPI-Proliferationsassay für drei humane kolorektale Adenokarzinomzelllinien (HCT116-luc, HT29-luc, SW620-luc) etabliert. Alternativ wurde der Effekt der 6 Substanzen auf die Koloniebildung im HTCA erfasst. Die Behandlung mit Chemerin-9 und Dexamethason führte zur signifikanten Zellzahlzunahme in allen drei Tumorzelllinien. GLP-2, Noradrenalin und Neurotensin steigerten die Tumorzellproliferation in maximal zwei Zelllinien. Dexamethason förderte auch die Koloniebildung bei allen drei Zelllinien, Chemerin-9, Noradrenalin und Neurotensin bei einer bzw. zwei Zelllinien. Die Vermittlung des in-vitro-Proliferationseffektes von Chemerin-9 über den G-Protein-gekoppelten Chemerinrezeptor CMKLR1 wurde mithilfe des ITCC-Internalisierungsassays verifiziert. Es wurden in den 5 von insgesamt 12 untersuchten Adenokarzinomzelllinien (SW480, HCT116-luc, HT29-luc, Colo320, LS174T) und einer Neuroblastomzelllinie (SK-N-AS) die Bindung und nachfolgende Internalisierung der ITCC-Fluoreszenzfarbstoff-markierten Chemerin-9-Variante nachgewiesen. Im Verdrängungsversuch zum Spezifitätsnachweis von der über den CMKLR1 vermittelten Chemerin-9-Internalisierung wurde eine geringgradige Signalabschwächung in den SW480, HT29-luc und SK-N-AS beobachtet. Zusätzlich wurde die Chemerin-9-Aufnahme in die subkutanen Tumore tragende Nacktmäuse mithilfe der in-vivo-Nahinfrarotbildgebung dargestellt. Der anschließende Behandlungsversuch mit der stabilen Chemerin-9-Variante demonstrierte ein Ansteigen des Tumorvolumens und -gewichtes in den HCT116-luc- und HT29-luc- Tumoren. Die vorliegende Arbeit lieferte einen ersten Hinweis auf die Bedeutung von Chemerin-9 als Wachstumsfaktor für das kolorektale Tumorwachstum in vitro und in vivo und brachte damit Anhaltspunkte für Strategien zur Therapie des kolorektalen Karzinoms mithilfe von Chemerinrezeptor- Antagonisten.","The growth of colon tumors and development of metastases are regulated by classic growth factors. The role of hormones like stress hormones and gastrointestinal peptides in this process is currently under investigation und represents the subject of this work, in which chemerin takes a key role. The plasma protein chemerin and its derivate chemerin-9 have different functions in the human organism. As a chemokine chemerin is involved among other things in inflammation process and induces adipo- and angiogenesis. The link between chemerin and colon cancer genesis has not been characterized, so far. In the present study, we studied the effect of the stress hormones dexamethasone and norepinephrine and the peptides neurotensin, GLP-2, somatostatin-analogue SOM230 and chemerin-9 on tumor cell proliferation in a screening approach. For that purpose we established a DAPI-proliferation assay for three human colon cancer cell lines (HCT116-luc, HT29-luc, SW620-luc). Alternatively we recorded the effect of these 6 substances on formation of tumor colonies in a HTCA. The treatment with chemerin-9 and dexamethasone led to significant increase of the cell number in all three cancer cell lines. GLP-2, norepinephrine and neurotensin raised cancer cell proliferation in maximal two cell lines. Dexamethasone promoted also colony formation in all three cell lines, chemerin-9, norepinephrine and neurotensin in one or two cell lines. The effect of chemerin-9 on tumor growth in vitro through the G protein-coupled receptor, CMKLR1, was verified with the help of ITCC-internalization assay. We detected binding and following internalization of chemerin-9 variant marked with ITCC-fluorescence dye in 5 of 12 colon cancer cell lines (SW 480, HCT116-luc, HT29-luc, Colo320, LS174T) and one neuroblastoma cell line (SK-N-AS). In competitive experiments a small signal reduction was observed in SW480, HT29-luc and SK-N-AS referring to the specifity of chemerin-9 internalization translated through CMKLR1. In addition we showed chemerin-9 accumulation in nude mice bearing subcutaneous tumors by in vivo near infrared imaging. A following treatment study with stable chemerin-9 variant demonstrated an increase of tumor volume and weight in HCT116-luc and HT29-luc tumors. The present study provides the first data of the importance of chemerin-9 as a potent growth factor for the colon cancer growth in vitro and in vivo.
