Sie dient dem besseren Verständnis der geometrischen Strukturen, ihrer Veränderungen sowie der grundlegenden Wechselwirkungen der ersten Reaktionsschritte und der Signalweiterleitung von zyklischen und linearen Tetrapyrrolen. Zu diesem Zweck wird eine modifizierte Methode der ultraschnellen polarisationsaufgelösten Infrarotspektroskopie vorgestellt, deren Verwendung die Analyse des Grundzustandes, des elektronisch angeregten Zustandes sowie der ersten Produktzustände der Photorezeptoren und Pigmente in struktureller sowie dynamischer Hinsicht erlaubt. So können z. B. relative Winkel zwischen den angeregten elektronischen Übergangsdipolmomenten und den abgetasteten Übergangsdipolmomenten der molekularen Schwingungen bestimmt werden. Da diese direkt mit der Konformation der Chromophore assoziiert sind, erhält man durch Kombinationen der bestimmten relativen Winkel mit Dichtefunktionaltheorie-Berechnungen Zugriff auf weitergehendes geometrisches und dynamisches Wissen, welches mit der herkömmlichen Infrarotspektroskopie nicht zugänglich ist. Der hier realisierte experimentelle Ansatz fand seine Anwendung in der Analyse verschiedener Tetrapyrrole sowie dem Laserfarbstoff Coumarin 314, der eine detaillierte Charakterisierung ermöglichte. Als bedeutendster Vertreter der zyklischen Tetrapyrrole wurde das Chlorophyll a (Chl a) untersucht, ein Pigment, dem eine wesentliche Rolle beim primären Energie- und Elektronentransfer der Photosynthese zukommt. Eine Voraussetzung für das Verständnis der Chlorophyll-Chlorophyll-Wechselwirkungen ist das Wissen der exakten Orientierung des elektronischen Übergangsdipolmoments μel der Qy-Bande innerhalb der Struktur des Chl a. Sie wurde erstmalig dreidimensional in gelöster Form der Pigmente bestimmt zu: μel = (0, 19/0, 98/0, 07). μel weicht um (12 ± 3)° von der Y-Achse ab und ragt um (4 ± 2)° aus der Ebene des Chlorinringes heraus. Die Eigenrotation der Chl a-Moleküle innerhalb des Lösungsmittels Toluol-d8 konnte mit einer Zeitkonstanten von t = (90 ± 10) ps beobachtet werden. Die Forschungen auf dem Gebiet der bakteriellen Phytochrome sind ein relativ neues Feld. Deren Chromophore, die linearen Tetrapyrrole, zeichnen sich durch eine reversible Photoisomerisation aus, wodurch die Phytochrome als Photorezeptoren viele biologische Prozesse steuern können. Das Phytochrom Agp1 des Agrobacteriums tumefaciens (Wildtyp sowie sterisch gelockt) wurde in dieser Arbeit analysiert. Es konnte eine spektrale Heterogenität des Absorptionsspektrums von Agp1 zweifelsfrei belegt und erstmalig der elektronische Übergang S0 -> S2 der Absorptionsbande bei 644 nm zugeordnet werden. Es wurde demonstriert, dass die wahrscheinliche Konformation des Chromophors im Grundzustand der Konformation ZZZssa entspricht und die Konformation ZZZasa ausgeschlossen werden kann. Eine elektronische Anregung des Chromophors führt ferner zu einer bi-exponentiellen Dynamik. Ein Teil der Anregungsenergie wird mit einer Zeitkonstanten von t = 5, 5 ps (mit Fehlerbereich von 4 bis 8 ps) durch lokal begrenzte Schwingungsrelaxation am Ring D im elektronisch angeregten Zustand abgeleitet.","This work is embedded in the framework of the Sonderforschungsbereich 498 ‘Protein-Cofactor Interactions in Biological Processes’ as part of the project B11 ‘Analysis of lightinduced vibrational structures of photoreceptors and antenna pigments’. It is focused on a better understanding of the geometrical structures, their changes and the fundamental interactions of the first reaction steps and the signal transduction of cyclic and linear tetrapyrroles. For this purpose, a modified ultrafast polarization-resolved infrared spectroscopy has been implemented. This experimental method allows the analysis of the ground state, electronically excited states and initial product states of the photoreceptors and pigments in terms of structural and dynamic aspects. Thus, it can for example determine relative angles between excited electronic transition dipole moments and probed transition dipole moments of the molecular vibrations. The transition dipole moments are directly associated with the conformation of the chromophores. Hence, a combination of the relative angles with density functional theory calculations provides access to geometric and dynamic knowledge that would not be accessible to conventional infrared spectroscopy. The presented experimental approach found its application in the analysis of different tetrapyrroles and the laser dye coumarin 314, which enabled a detailed characterization. As the most important representative of the cyclic tetrapyrroles chlorophyll a (Chl a) was analyzed, a pigment that plays an essential role in the primary energy and electron transfer in photosynthesis. A prerequisite for the understanding of chlorophyllchlorophyll interactions is the knowledge of the exact orientation of the electronic transition dipole moment μel of the Qy-band within the structure of the Chl a. For the first time, μel was determined in three dimensions of the pigments in soluted form: μel = (0.19/0.98/0.07). μel differs by (12 ± 3)° from the Y-axis and rises by (4 ± 2)° out of the chlorin plane. The rotation of the Chl a molecules in the solvent toluene-d8 was observed with a time constant of t = (90 ± 10) ps. The research in the field of bacterial phytochromes is a relatively new field. Their chromophores, linear tetrapyrroles, are characterized by a reversible photoisomerization, by which the phytochromes control many biological processes as photoreceptors. The phytochrome Agp1 of Agrobacterium tumefaciens (wild type and locked) was analyzed in this work. The spectral heterogeneity of the absorption spectrum of Agp1 was proven unambiguously and the absorption band at 644 nm was assigned to the electronic S0 -> S2 transition for the first time. Furthermore, it was demonstrated that the most probable conformation of the chromophore in the ground state is the ZZZssa conformation and that the ZZZasa conformation can be excluded. An electronic excitation of the chromophore also results in a bi-exponential dynamic.
