Deswegen scheint dieses Eicosanoid eine besondere Rolle bei der Rekrutierung von PMN in Entzündundsreaktionen zu spielen. PMN besitzen eine Vielzahl von Rezeptoren, die am Priming und an der Zellaktivität beteiligt sind. Unter Priming versteht man eine Konditionierung der Zellen, durch die ein anderer Stimulus eine verstärkte Aktivität der Zellen hervorrufen kann. Einer von diesen Rezeptoren ist der Formyltripeptidrezeptor (FPR). Er vermittelt sowohl Chemotaxis als auch die Aktivierung des Nicotinamidadenindinukleotidphosphat-Oxidasesystems (NADPH- Oxidasesystems). Es ist bekannt, daß hierbei die LTB4?Synthese nur wenig angeregt wird, obwohl sowohl Arachidonsäure (AA) freigesetzt als auch die intrazelluläre Kalziumkonzentration (Cai2+) stark erhöht wird, beides Faktoren, die für die LTB4-Formation gebraucht werden. Um diesen Widerspruch aufzuklären, wurden die Freisetzung von AA und die LTB4-Synthese in mit 14C-AA markierten PMN unter Verwendung von verschiedenen Inhibitoren des Phospholipidstoffwechsels untersucht. Die Ergebnisse zeigten, daß der größte Anteil der AA-Freisetzung durch die cytosolische Phospholipase A2 Gruppe IVa (cPLA2) und die Phospholipase D (PLD) ausgelöst wurde, während die sekretorische Phospholipase A2 (sPLA2) keine Rolle zu spielen scheint. Durch die Hemmung der Phosphohydrolase der Phosphatidsäure (PA) mit Propranolol kam es zum Abfall der AA-Freisetzung, jedoch zum Anstieg der 5-Lipoxygenase-Aktivität (5-LOX?Aktivität). Eine Inhibition der NADPH-Oxidase-Aktivität verursachte einen leichten Anstieg der 5-LOX-Produkte. Die Länge und Stärke des Kalziumsignals sind scheinbar die kausalen Faktoren für die fMLP-induzierte LTB4-Synthese. Weder die freie AA noch die enzymatische Aktivität der 5-LOX sind die limitierenden Faktoren für die fMLP induzierte Produktion des LTB4.","Leukotriene B4 (LTB4) is an important proinflammatory mediator which is produced by neutrophils (PMN) and exhibits potent chemotactic activity towards various inflammatory cells. Therefore this eicosanoid is believed to play a central role in the recruitment of PMN at inflammatory sites. PMN posesses a number of receptors that are essential for priming and activation of their functions (Priming means a treatment that does not itself elicits a response in a cellsystem but induces an enhanced capacity of a cell to respond to a second stiumulus). One of the receptors is the formyl tripeptide receptor (FPR) the agonists of which cause both chemotaxis and activation of the NADPH oxidase system. However, formyl peptides such as formyl-methionyl-leucyl- phenylalanine (fMLP) are known to be inducers of LTB4-synthesis in human PMN, albeit they cause both rise of cytosolic calcium concentration and liberation of arachidonic acid (AA). Both of them are required for LTB4 formation. To elucidate this discrepancy, we investigated AA-release and LTB4 synthesis from 14C-AA prelabelled cells by the use of various inhibitors of the phospholipid metabolism. From our results we conclude that the major part of AA-release is triggered by cytosolic phospholipase A2 (cPLA2) and phospholipase D (PLD) pathways without any involvement of secretory phospholipases A2 (sPLA2). The blockade of phosphatidic acid (PA) phosphohydrolase by propranolol in the PLD pathway leads to a decrease in AA-release and concomitantly to a significant increase in LTB4?synthesis. Inhibition of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase activity caused only slight enhancement of the LTB4 formation.
