Die Materialien zeigten im Test nach ISO 10993-5 mit 3T3 Mausfibroblasten keine zytotoxische Wirkung. Die Untersuchungen mit HUVECs hinsichtlich der Zell-Material- Wechselwirkung (Adhäsion, Proliferation, Funktion) belegten eine gute Biokompatibilität für PPDO/Diorez. Während die Zellzahl auf PPDO/Diorez 50/50 gegenüber PP insgesamt höher war und im Laufe der Kultivierung anstieg, blieb diese bei PPDO/PCL 50/50 und PPDO/PCL 40/60 weitgehend konstant. Auf den PPDO/PCL–Materialien kam es demzufolge zu einer geringen Adhäsion von HUVEC. Mit Hilfe von Oberflächenbeschichtungen durch Zelladhäsionsproteine wie Fibronektin, Fibrinogen oder Collagen I konnte die Zelladhäsion und -proliferation auch auf den PPDO/PCL–Polymeren gesteigert werden. Morphologisch zeigten sich bei Anfärbung von Aktin in Abhängigkeit vom Material Unterschiede. Die Funktionalität der Zellen in Bezug auf ihre antikoagulante Wirkung war auf jedem Material vorhanden. Die betrachteten bioabbaubaren Polymere haben sich in den durchgeführten in vitro Versuchen somit als biokompatibel und damit als vielversprechende Materialien für eine Anwendung als Implantatmaterial erwiesen. Für spezielle Applikationen ist eine Oberflächenmodifikation mit adhäsionsfördernden Substanzen notwendig. Bei einem möglichen in vivo Einsatz könnten sie durch eine optimale Festigkeit des Knotens und einer einstellbaren Resorptionszeit zu einer guten Wundheilung und damit einer verbesserten postoperativen Kreislaufstabilisierung und damit einer kürzeren Genesungszeit beitragen. Dabei muss beachtet werden, dass es sich bei den durchgeführten Versuchen um in vitro Untersuchungen handelt. Im Organismus hängt die Gewebebiokompatibilität allerdings von komplexeren Wechselwirkungen ab, die Gegenstand weiterer Untersuchungen sein müssen.","The study presented here was able to demonstrate that all the tested polymers are biocompatible materials. The materials showed no cytotoxic effect in the tests according to EN ISO 19993-5 with 3T3 mouse fibroblasts. The investigations with HUVEC with regard to the interactions between cells and materials (adhesion, proliferation, function) confirm a good biocompatibility for PPDO/Diorez. While the total number of cells was higher on PPDO/Diorez 50/50 compared to PP and increased in the course of cultivation, it remained largely constant on PPDO/PCL 50/50 and PPDO/PCL 40/60. The adhesion of HUVEC on the PPDO/PCL materials was thus marginal. With the aid of surface coatings with cell adhesion proteins like fibronectin, fibrinogen or collagen I, cell adhesion and proliferation on the PPDO/PCL polymers could be increased as well. Morphologically, material-dependent differences were observed with actin staining. The functionality of the cells with regard to their anticoagulant effect was seen on all the materials. The polymers examined in the in vitro trials thus proved to be biocompatible and are therefore very promising materials for use as implant material. Special applications necessitate a surface modification with adhesion-promoting substances. In in vivo applications, these materials can contribute to good wound healing and thus to an improvement of postoperative circulatory stabilization as well as shorter recovery times because of optimal knot tightness and an adjustable adsorption time. In this context, attention must be paid to the fact that the trials carried out here are in vitro examinations.
