Die Sialinsäuren, als endständige Komponenten der Oligosaccharidketten sind in eine Vielzahl von zellulären Interaktionen wie Zell-Zell-Adhäsion, Zellmigration und Metastasierung involviert. Sie dienen weiterhin als Erkennungsdeterminanten für Pathogene und tragen zur Stabilität von Glycoproteinen bei. Die N-Acetylneuraminsäure ist der biologische Vorläufer aller natürlich vorkommender Sialinsäuren. Die ersten Schritte der Sialinsäurebiosynthese, die Epimerisierung von UDP-GlcNAc zu ManNAc und die anschließende Phosphorylierung in der C-6-Position werden von der UDP-N-Acetylglucosamin-2-Epimerase/N-Acetylmannosaminkinase (GNE), dem Schlüsselenzym der Sialinsäurebiosynthese, katalysiert. Im Rahmen dieser Arbeit wurden zwei neue Isoformen der hGNE, hGNE2 und hGNE3, identifiziert. Im Gegensatz zu hGNE1 besitzt hGNE2 einen verlängerten, hGNE3 einen verkürzten N-Terminus. Während GNE2 auch in anderen Spezies wie Affe, Maus, Ratte, Huhn oder Fisch gefunden wurde, beschränkt sich das Vorkommen von GNE3 auf die Primaten. Sowohl die humanen als auch die murinen Isoformen zeigen gewebsspezifische Expressionsmuster. hGNE1, hGNE2, mGNE1 und mGNE2 wurden über das BAC-TO-BAC®-Baculovirus-System funktionell exprimiert und charakterisiert. Die His-getagten Fusionsproteine konnten als lösliche aktive Enzyme exprimiert werden und mittels Affinitätschromatographie in mg-Mengen gewonnen werden. Das hGNE3-Protein konnte in E.coli BL21-Zellen funktionell, aber nur in geringen Mengen exprimiert werden. Während alle rekombinanten Proteine ManNAc-Kinase- Aktivität besaßen, bildeten nur hGNE1, mGNE1 und mGNE2 UDP-GlcNAc-2-Epimerase- aktive Tetramere. Bei hGNE2 war die UDP-GlcNAc-2-Epimerase-Aktivität um den Faktor 5 reduziert, was auf die Bildung von Dimeren zurückzuführen war. Für hGNE3 konnte gezeigt werden, daß das Enzym keine Epimeraseaktivität mehr besaß. Im letzten Teil der Arbeit wurden die Proteine VCP und Oxr1 auf mögliche Interaktionen mit der GNE hin untersucht. Die Ergebnisse implizieren, daß sowohl VCP als auch Oxr1 keine Interaktionspartner der GNE sind.","Sialylation of glycoproteins and glycolipids on cell surfaces has an important role during development, differentiation and inflammation, as well as in the pathogenesis of diseases. Sialic acids are terminal components of oligosaccharides and involved in a variety of cellular interactions, such as cell-cell adhesion, cell migration and metastasis. They are also known to be involved in the formation of recognition determinants of pathogens and stability of glycoproteins. The N-acetylneuraminic acid is the biological precursor of all naturally occurring sialic acids. The first two steps, the epimerization of UDP-GlcNAc to ManNAc and the consecutive phosphorylation at C-6, are catalyzed by the bifunctional enzyme UDP-N-acetylglucosamine 2-epimerase/N-acetylmannosamine kinase (GNE), the key enzyme of the sialic acid biosynthesis. In this thesis two novel isoforms of human GNE, hGNE2 and hGNE3, have been identified. Opposed to hGNE1, hGNE2 posseses an extended and hGNE3 has a deleted N-terminus, respectively. GNE2 was also found in other species like apes, mouse, rat, chicken and fish, whereas GNE3 seems to be restricted to primates. Human and mouse isoforms displayed tissue specific expression pattern. hGNE1, hGNE2, mGNE1 and mGNE2 were functionally expressed by the BAC-TO-BAC® baculovirus system. The 6xhis-tagged fusion proteins could be expressed as soluble active enzymes and purified by affinity chromatography in mg amounts. The hGNE3 protein was functionally expressed in E.coli BL21 cells, but its amount was very low. All recombinant proteins displayed ManNAc kinase activity. hGNE1, mGNE1 and mGNE2 formed UDP-GlcNAc 2-epimerase active tetramers. hGNE2 showed a 5-fold reduced UDP-GlcNAc 2-epimerase activity, which was due to the formation of dimers. hGNE3 displayed no epimerase activity at all. In the last part of this thesis the proteins VCP and Oxr1 were analyzed for potential interactions with the GNE protein.
