Dies geschieht wahrscheinlich über einen PI3-K, PKA und Akt- abhängigen Signalweg, der zu einer Phosphorylierung der eNOS an Ser1179 führt. Zunächst konnte an Rattenaortenringen eine EGCG-induzierte, dosisabhängige Vasorelaxation nachgewiesen werden, die bis zu einer EGCG-Konzentration von 25 µM endothel- und NO-abhängig war. Die Stimulation zweier verschiedener Endothelzelltypen mit EGCG führte zu einer dosisabhängigen Aktivierung der eNOS, was durch einen eNOS-Aktivitätstest in lebenden Zellen nachgewiesen werden konnte. Durch Western Blots konnte gezeigt werden, dass bei dieser schnellen eNOS-Aktivierung eine Phosphorylierung des Enzyms involviert ist. Eine 15-minütige Inkubation mit 100 µM EGCG führte zu einer Phosphorylierung von eNOS an Ser1179, von Akt an Ser473 und von ERK1/2 an Thr202/Tyr204. Die Aktivierung der eNOS und Phosphorylierung von Ser1179 wurden durch 30-minütige Vorinkubation der Zellen mit PI3-K-Inhibitoren (LY 294002, Wortmannin) vollständig gehemmt. Inkubation mit Inhibitoren der PKA (PKI) oder der Akt (SH-5) senkten die EGCG-induzierte eNOS-Aktivierung partiell. SH-5 verhinderte sowohl die Phosphorylierung der Akt an Ser473, als auch die Phosphorylierung der eNOS an Ser1179. Obwohl PKI keinen Einfluss auf die Phosphorylierung dieser Stellen hatte, hemmte es die Erhöhung der Aktivität des Enzyms durch EGCG. Im PKA-Assay konnte eine EGCG-induzierte Aktivierung der PKA nachgewiesen werden. PD98059, ein Inhibitor der ERK1/2, hatte keinen Einfluss auf die Aktivierung und die Ser1179-Phosphorylierung der eNOS. Eine Behandlung von BAECs und CPAEs mit verschiedenen EGCG-Konzentrationen über 24 beziehungsweise 72 h hatte keine Auswirkungen auf die eNOS-Proteinmenge in der Zelle. Diese Ergebnisse legen den Schluss nahe, dass die schnelle Aktivierung der eNOS durch EGCG durch eine Phosphorylierung von eNOS an Ser1179 bedingt ist. Offensichtlich sind mehrere Kinasen involviert, die möglicherweise parallel an verschiedenen eNOS-Phosphorylierungsstellen wirken. Welche Kinasen letztendlich welche Aminosäuren der eNOS phosphorylieren, muss in weiteren Versuchen geklärt werden.","In the present study, we provide the first experimental evidence that the tea catechin EGCG induces an activation of the endothelial NO-synthase in endothelial cells, probably via a PI3-K-, PKA- and Akt-dependent pathway, that leads to phosphorylation of eNOS at Ser1179. We observed an EGCG-induced, dose-dependent vasodilation of the rat aorta, which remained endothelium- dependent up to an EGCG-dose of 25 µM. Stimulation of two different types of endothelial cells lead to dose-dependent activation of eNOS. The western blot analysis showes, that 15 min stimulation of bovine aortic endothelial cells with 100 µM of EGCG leads to phosphorylation of eNOS at Ser1179, Akt at 473 and ERK1/2 at Thr202/Tyr204. Activation of eNOS and phosphorylation at Ser1179 were completely inhibited by previous incubation of the cells with PI3-K-inhibitors (LY294004, wortmannin). Incubation with inhibitors of PKA (PKI) or Akt (SH-5) decreased EGCG-induced eNOS-activation partially. SH-5 prevented phosphorylation of Akt at Ser473 as well as phosphorylation of eNOS at Ser1179. PKI had no influence on the phosphorylation of these sides, but it inhibited EGCG-induced increase of eNOS-activity. PD98059, an inhibitor of ERK1/2, did not affect EGCG-induced activity of eNOS and phosphorylation at Ser1179. Treatment of BAECs and CPAEs with different concentrations of EGCG for 24 or 72 hours had no influence on the content of eNOS-protein in these cells. These results suggest that rapid EGCG-induced eNOS-activation is conditioned by a phosphorylation of eNOS at Ser1179. Obviously there are different kinases involved, which act parallel on different phosphorylation- sites of eNOS.
