Rolf Bräuer","w","2011-06-05","2018-06-07T19:58:33Z","2011-05-30T08:52:36.305Z","2011","Bei der G6PI-induzierten Arthritis entwickeln DBA1-Mäuse nach Immunisierung mit G6PI eine akute, symmetrische Polyarthritis, die im Gegensatz zu dem T-Zellrezeptor transgenen K/BxN-Modell nicht durch Transfer von Antikörpern übertragen werden kann. Damit konnte zum ersten Mal gezeigt werden, dass eine Immunisierung mit einem systemischen Antigen eine organspezifische Erkrankung in nicht transgenen Mäusen induziert. Da in der G6PI-induzierten Arthritis die Arthritisentwicklung entscheidend von Fc-Rezeptoren abhängt, wurde vermutet, dass auch B-Zellen und die von ihnen produzierten anti-G6PI-Antikörper eine pathogene Rolle spielen. Im ersten Teil der Arbeit wurde die Kinetik und Lokalisation der B-Zellantwort gegen G6PI nach Immunisierung mit rhG6PI in DBA/1-Mäusen untersucht. Anders als im K/BxN-Modell wird die B-Zellantwort hauptsächlich in den die Injektionsstellen drainierenden Lymphknoten initiiert. In dieser frühen Phase der Immunantwort entsteht eine große Anzahl von kurzlebigen Plasmazellen, die nach wenigen Tagen durch Apoptose noch vor Ort sterben. Gleichzeitig wandert ein Teil dieser Zellen in das Knochenmark, wo sie zu langlebigen Plasmazellen differenzieren, um dort auch nach Abklingen der klinischen Symptomatik hohe Antikörpertiter gegen G6PI zu produzieren. Für die Arthritisentwicklung scheint es von Bedeutung zu sein, welche Epitope der G6PI von den anti-G6PI-Antikörpern erkannt werden. Im zweiten Teil der Arbeit wurden daher die qualitativen Eigenschaften der polyklonalen anti-G6PI- Antikörper analysiert. Mit Hilfe des Pepscan-Verfahrens wurden die anti-G6PI- Antikörper suszeptibler und nicht suszeptibler Stämme auf ihre Epitopspezifität untersucht und miteinander verglichen. Hauptepitop der G6PI- spezifischen Antikörper ist sowohl bei den suszeptiblen als auch nicht suszeptiblen Stämmen das Epitop rhG6PI458-472 mit der Aminosäuresequenz DLERLLPHKVFEGNR, womit die pathogene Bedeutung dieses Epitops zumindest als alleiniges arthritogenes Epitop ausgeschlossen werden kann. Es ist aber möglich, dass es bei den DBA/1-Mäusen in Kombination mit anderen Epitopen, für die Arthritisentwicklung eine Bedeutung haben könnte. Die Immunisierung mit dem Epitop rhG6PI458-472 führte weder klinisch noch histologisch zur Entwicklung einer Arthritis. Auch zeigten die CD4+-T-Zellen in den DBA/1-Mäusen nach Restimulation mit dem identifizierten B-Zellepitop rhG6PI458-472 keine Proliferation. Diese Ergebnisse sprechen gegen die Induzierbarkeit der G6PI-induzierten Arthritis durch anti-G6PI-Antikörper als alleinigen Faktor, jedoch nicht gegen deren generelle pathogene Bedeutung. Die Generierung und Charakterisierung monoklonaler anti-G6PI-Antikörper aus arthritischen Mäusen sowie Transferversuche, bei denen mehrere monoklonale anti-G6PI-Antikörper miteinander kombiniert werden, könnten weiteren Aufschluss darüber geben, ob bisher die Quantität oder die Qualität der transferierten Antikörper nicht hinreichend für eine Arthritisinduktion waren.","Immunization of DBA/1 mice with recombinant human G6PI (rhG6PI) leads to acute symmetrical peripheral polyarthritis of the small distal joints. Thus, the G6PI-induced arthritis demonstrates the induction of organ-specific disease induced by systemic autoimmunity in genetically unaltered mice. In contrast to the TCR transgenic K/BxN model, transfer of serum or purified IgG from arthritic DBA/1 mice does not induce arthritis in naive recipients. Nonetheless, B cells and antibodies are necessary for G6PI-induced arthritis since Fc[gamma]R+ effector cells are crucial for disease induction. As a first step in identifying the role of B cells as antibody-secreting cells in the G6PI-induced arthritis model, anti-G6PI B cell response was measured before, while and after clinical disease. Unlike the K/BxN model, B cell response against the ubiquitously expressed G6PI was initiated in and focused to the lymph nodes draining the immunization site. In the early phase of the humoral immune response a large amount of plasma cells is generated. Most of these cells are short-lived plasma cells, which die within few days in this location by apoptosis. However, some plasma cells leave the lymph nodes and migrate to the bone marrow where they differentiate into long-lived plasma-cells to provide persistent high antibody titer against G6PI, although clinical disease has already resolved. The arthritogenicity of antibodies might depend on their epitope-specificity against G6PI. In the second part of this study the quality characterristics of anti-G6PI antibodies were analyzed. Following the pepscan technique linear epitopes of anti-G6PI antibodies from susceptible and non- susceptible strains were identified. Our findings suggest that the epitope rhG6PI458-472 with the amino acid sequence DLERLLPHKVFEGNR is the main epitope detected by anti-G6PI antibodies from both suscepitble and non-susceptible strains. Thus, the arthritogenicity of the anti-G6PI antibodies does not exclusively depend on the recognition of rhG6PI458-472, but it is still possible that in DBA/1 mice a combination of antibodies binding to rhG6PI458-472 and other different epitopes is required to induce arthritis. Immunization with rhG6PI458-472 did not lead to clinical and histological disease. Additionally no cellular response against rhG6PI458-472 was observed after immunization with rhG6PI. In summery, the above findings indicate that antibodies are necessary, but not sufficient alone for G6PI-induced arthritis.
