Olze","n","2009-09-18","2018-06-07T19:23:57Z","2009-09-24T11:59:30.915Z","2009","It has been known that inflammation and inflammatory cytokines are involved in pathogenesis of many types of cochlear disorders including ototoxicity. Stress proteins such as HSP70 are induced in response to various stresses in the inner ear and presents otopretective effects. My dissertation was dedicated to study ototoxic and otoprotective properties of selected substances in the inner ear cells and tissues. In the first part of my work, I studied the gene expression profile of inflammatory cytokines, HSP70 and prestin in the organ of Corti during organotypic culture. I found that exposure to salicylate (2.5 mM) upregulated the number of transcripts encoding IL-6, HSP70 and prestin in the organ of Corti. These studies were complemented by expression study using an auditory epithelium cell line, which yielded similar results. The overall results suggested that salicylate may work as an ototoxin not only by directly influencing motility of outer hair cells (as previously described) but also by affecting gene expression. Interestingly, changes in transcription detected by quantitative real time RT-PCR were not followed by changes in translation, as per Western blot or ELISA of IL-6 and HSP70. From the first part of my work, I knew that the transcription of IL-6 increases after exposure to salicylate in the organ of Corti during organotypic culture. In addition, the reports of others showed cochlear increase in IL-6 during trauma-, noise- or cisplatin- induced injury. That is why in the second part of my work, I studied the influence of exogenous IL-6 on ototoxicity in the organ of Corti explant cultures. I discovered that the exogenous IL-6 (up to 90 ng/ml) had no ototoxic properties when applied on its own, as determined by cochleogram. Moreover, when IL-6 (30 ng/ml) was added to OC explants together with cisplatin (15µM for 48 h), it had partially protected hair cells from cisplatin-induced ototoxicity. The mechanism of the otoprotective effect induced by IL-6 in auditory hair cells needs to be clarified. In the last part of my work, I studied otoprotection induced by chemical stimulators of heat shock protein 70. To date, resveratrol and geldanamycin were demonstrated to induce HSP70 production. Based on data showing unequivocally that intracellular endogenous HSP70 is otoprotective, I used the two substances mentioned above to induce HSP70 in the organ of Corti explant cultures. The induction of HSP70 by resveratrol or geldanamycin was analyzed by RT-PCR and ELISA. Resveratrol (up to 200 µM) has not induced HSP70 expression in the organ of Corti explant. In contrast, geldanamycin (2 µM) induced HSP70 expression in OC cultures at both mRNA and protein levels. Fluorescence microscopy revealed that HSP70 induced by geldanamycin was observed mainly in hair cells and interdental cells of spiral limbus, but not in either spiral ganglion cells or nerve fibers. Next, I studied the otoprotective properties of geldanamycin using a standard ototoxic antibiotic gentamicin (500 µM). In gentamicin-damaged OC explants, geldanamycin significantly reduced the loss of outer but not inner hair cells. The differential protective effect of geldanamycin between outer and inner hair cells indicated that HSP70-independent pathway may contribute to the protective effect of geldanamycin. I observed the otoprotective effect of geldanamycin in OC explants pretreated with geldanamycin and in those treated simultaneously with geldanamycin and gentamicin. Further experiments are needed to reveal the molecular mechanisms underlying the otoprotective role of geldanamycin, which may provide insights for therapeutic approaches aimed at preventing ototoxicity induced by various ototoxic agents.||Es ist bekannt, dass Entzündung und entzündliche Zytokine in die Pathogenese verschiedener kochleärer Erkrankungen einschließlich Ototoxizität involviert sind. Stressproteine wie HSP70 werden im Innenohr als Antwort auf unterschiedliche Stressoren induziert und haben otoprotektive Wirkungen. Im ersten Teil meiner Arbeit habe ich das Genexpressionsprofil von entzündlichen Zytokinen, HSP70 und Prestin im Corti-Organ in der organotypischen Kultur untersucht. Dabei fand ich heraus, dass Salizylat (2,5 mM) die Zahl der Transkripte, die IL-6, HSP70 und Prestin kodieren, im Corti-Organ erhöht. Diese Untersuchungen wurden durch Expressionsstudien, in denen eine auditorische Epithelzelllinie benutzt wurde, ergänzt und bestätigt. Diese Ergebnisse ließen vermuten, dass die ototoxische Wirkung von Salizylat nicht nur die Motilität der äußeren Haarzellen direkt beeinflusst, sondern auch die Genexpression verändert. Interessanterweise folgten die Veränderungen in der Translation von IL-6 und HSP70, mittels Western Blot oder ELISA bestimmt, nicht denen in der Transkription, die mittels quantitativer real time RT-PCR gefunden wurden. Wie aus dem ersten Teil meiner Arbeit hervorging, erhöhte sich die Transkription von IL-6 im Corti-Organ nach Salizylatzugabe während der organotypischen Kultur. Zusätzlich ist in der Literatur über eine kochleäre Erhöhung von IL-6 während eines durch Trauma, Lärm oder Cisplatin induzierten Schadens berichtet worden. Deshalb habe ich im zweiten Teil meiner Arbeit den Einfluss von exogenem IL-6 auf die Ototoxizität in der Explantkultur des Corti-Organs untersucht. Ich konnte mittels Kochleogramm nachweisen, dass exogenes IL-6 (bis zu 90 ng/ml) bei alleiniger Zugabe keine ototoxischen Eigenschaften hatte. Darüber hinaus reduzierte IL-6 (30 ng/ml) teilweise die durch Cisplatin induzierte Ototoxizität, wenn es zusammen mit Cisplatin (15 µM, 48 h) den Explantaten des Corti-Organs zugesetzt wurde. Der Mechanismus, der dieser otoprotektiven Wirkung von IL-6 in den auditorischen Haarzellen zugrunde liegt, muss noch geklärt werden. Im letzten Teil meiner Arbeit untersuchte ich die Otoprotektion durch chemische Stimulatoren von Hitzeschockprotein 70. Es wurde bereits nachgewiesen, dass Resveratrol und Geldanamycin die Produktion von HSP70 induzieren. Basierend auf Daten, die eindeutig gezeigt haben, dass intrazelluläres endogenes HSP70 otoprotektiv ist, benutzte ich die beiden o.g. Substanzen, um die HSP70-Produktion in den Explantkulturen des Corti-Organs hervorzurufen. Die Induzierung von HSP durch Resveratrol oder Geldanamycin wurde mittels RT-PCR und ELISA analysiert. Resveratrol (bis zu 200 µM) hat die HSP70-Expression in den Kuluren des Corti- Organs nicht hervorgerufen. Im Gegensatz dazu induzierte Geldanamycin (2 µM) die HSP70-Expression in den Kulturen des Corti-Organs sowohl auf mRNA- als auch auf Proteinebene. Fluoreszenzmikroskopische Untersuchungen ergaben, dass Geldanamycin HSP70 hauptsächlich in den Haarzellen und Interdentalzellen des Limbus spirale, aber nicht im Spiralganglion oder in den Nervenfasern induzierte. Anschließend untersuchte ich die otoprotektiven Eigenschaften von Geldanamycin, indem ich das Antibiotikum Gentamycin (500 µM), das bekannt für seine Ototoxizität ist, benutzte. In den durch Gentamycin geschädigten Explantaten des Corti-Organs reduzierte Geldanamycin den Verlust der äußeren, aber nicht der inneren Haarzellen signifikant. Der unterschiedliche protektive Effekt von Geldanamycin auf die inneren und äußeren Haarzellen lässt vermuten, dass ein von HSP70 unabhängiger Mechanismus zu dieser Protektion beitragen könnte. Weitere Experimente sind notwendig, um die molekularen Mechanismen, die der otoprotektiven Rolle von Geldanamycin zugrunde liegen, zu klären.
