Die Dissertation befasst sich mit dem Serumprofil des Lipoproteinstoffwechsel als wichtigem Indikator des gesamten menschlichen Metabolismus. Der Studie liegen klinisch relevante Phänotypen wie Gesamtcholesterin, LDL- und HDL-Cholesterin und Triglyzeride zugrunde. Als Genotyp wurden Single Nucleotide Polymorphisms (SNPs) sowie Haplotypen innerhalb von Kandidatengenen des Lipoproteinstoffwechsels analysiert. Zentrales Thema ist die statistische Analyse eines Datenpools, der wichtige epidemiologische, klinische, biochemische und genetische Merkmale einer umfassenden Stichprobe deutscher Familien und einer Schweizer Bevölkerungsstichprobe enthält. Gen-basierte Analyse und Replikation von Assoziationsbefunden der Kandidatenloci stehen im Mittelpunkt der Arbeit. Basierend auf einem Modell multipler genetischer Marker konnte ein maßgeblicher Anteil genetisch bedingter phänotypischer Varianz für LDL, HDL und den Quotienten LDL/HDL in der allgemeinen deutschen Bevölkerung erklärt werden. Diese Befunde wurden an einer unabhängigen Stichprobe validiert. Die Replikationsstudie zeigte signifikante Assoziation für 5 der 10 untersuchten Gen-Loci mit der Variation von LDL- und HDL-Cholesterin.","Familiar background is one of the most important risk factors of complex diseases like cardiovascular disease, cancer, diabetes mellitus, etc. The identification of underlying genetic influences is an important step towards revention, diagnosis and medication. The dissertation is concerned with the serumprofile of lipid metabolism as an important indicator of the human metabolism in general. The clinically relevant phenotypes total cholesterol, LDL- and HDL- cholesterol and triglycerides form the basis of the study. Single Nucleotide Polymorphisms (SNPs) and haplotypes in candidate genes of lipid metabolism are analysed as genotype. Central issue is the statistical analysis of a data-pool containing important epidemiological, clinical, biochemical and genetic parameters of a sample of nuclear families from Germany and a population based sample from Switzerland. Gen-based analysis and replication of association findings in candidate regions are focused. Results show that models of multiple genetic markers explain most of the genetic variance in LDL, HDL, and LDL/HDL-ratio in the German sample. These findings are validated in an independent sample.
