Deshalb ist es für das bessere Verständnis und zukünftige Behandlungen wichtig, Veränderungen im Glutamat-System im ZNS und die damit verbundenen Risikogene für endogene Psychosen zu erforschen und besser zu verstehen. Gegenstand dieser Dissertation war die Untersuchung von Konzentrationen von Glutamat mittels Protonen-Magnet-Resonanz-Spektroskopie (1H-MRS) im anterioren zingulären Kortex und im Hippocampus und die Einflüsse des DTNPB1-Gens, das mit Störungen im Glutamat-System in Verbindung gebracht wird. In einer ersten Studie wurden dabei signifikant veränderte Glutamat-Konzentrationen im anterioren zingulären Kortex und im Hippocampus von Patienten mit bipolar affektiver Störung gefunden. In einer zweiten Studie wurde in gesunden Probanden die Verbindung zwischen genetischen Varianten des DTNBP1-Gens und der Glutamat-Konzentration im anterioren zingulären Kortex und Hippocampus untersucht. Ergänzt wird die Dissertation durch eine klinische Studie zur Untersuchung von sozialer Kognition am Beispiel der ‚Theory of Mind’ (ToM) bei Patienten mit bipolar affektiver Störung.","Glutamate as the major excitatory neurotransmitter in the central nervous system plays a key role in the genesis of endogenous psychosis like schizophrenia and bipolar affective disorder. Therefore, researches on the changes of the glutamate system and underlying candidates of risk genes for endogenous psychosis seem to be of importance for a better understanding and future treatment. In this dissertation, we examined concentrations of glutamate using proton magnetic resonance spectroscopy (1H-MRS) in the anterior cingulate cortex and hippocampus and the impact of DTNBP1-gene associated to the glutamate system. In a first study significant changes in glutamate concentrations in the anterior cingulate cortex and hippocampus of patients with bipolar disorder were found. In a second study of healthy subjects, an association between genetic variants of the DTNBP1-gene and glutamate concentrations in the anterior cingulate cortex and hippocampus was investigated.
