Kreis","m","2013-02-01","2018-06-07T19:01:14Z","2013-01-16T13:36:40.640Z","2013","Das in den X/A-ähnlichen Zellen des Magens synthetisierte Peptidhormon Ghrelin und sein Metabolit Desacyl-Ghrelin sind an der Regulation der Nahrungsaufnahme in Menschen und Nagern beteiligt. Ghrelin induziert nach intraperitonealer Injektion sowohl einen Anstieg der Nahrungsaufnahme wie auch einen signifikanten Anstieg der Anzahl der c-Fos-positiven Neurone als Marker für neuronale Aktivierung in drei hypothalamischen Nuclei. Im Einzelnen sind dies der Nucleus arcuatus, der dorsomediale Hypothalamus und der Nucleus paraventricularis. Die Ghrelin-Effekte auf die Nahrungsaufnahme werden über Neuropeptid Y-/Agouti related peptide-immunreaktive Neurone vermittelt. Eine simultane Injektion von Desacyl-Ghrelin, welches sich von Ghrelin durch das Fehlen einer Octanoyl-Gruppe unterscheidet, und Ghrelin bewirkt eine Blockade des Ghrelin-Effekts auf die Nahrungsaufnahme. Zusätzlich konnte beobachtet werden, dass die Anzahl c-Fos-positiver Neurone im Nucleus arcuatus nach simultaner Injektion der Peptide signifikant erniedrigt ist im Vergleich zur alleinigen Ghrelin-Injektion. Die singuläre Injektion von Desacyl-Ghrelin hat keine Suppression der Nahrungsaufnahme zur Folge. Etwa die Hälfte der aktiven Neurone nach simultaner Injektion der beiden Peptide ließ sich mittels immunhistochemischer Doppelfärbungen als Nesfatin-1-positiv darstellen. Das anorexinogen wirksame Peptid Nesfatin-1 ist hierbei vermutlich an der Vermittlung des inhibitorischen Effekts von Desacyl-Ghrelin auf die Ghrelin- induzierte Nahrungsaufnahme beteiligt. Die Serin-Threonin-Kinase mammalian target of Rapamycin lässt sich im Hypothalamus in Neuronen des Nucleus arcuatus nachweisen. Eine erhöhte Konzentration der aktivierten, phosphorylierten Form dieses Proteins im Nucleus arcuatus unterdrückt die Nahrungsaufnahme. Das Neuropeptid cocaine and amphetamine regulated transcript ist ebenfalls an der Regulation der Nahrungsaufnahme beteiligt. Es bewirkt nach peripherer Injektion, genau wie Nesfatin-1, eine Inhibition der Nahrungsaufnahme. In einer Studie dieser Forschungsarbeit konnte durch Doppelfärbungen gezeigt werden, dass die genannten Peptide intrazellulär kolokalisiert sind. Denkbar sind hier sowohl modulatorische als auch additive Effekte im Zusammenspiel dieser anorexinogenen neuronalen Hormone.","Ghrelin and its metabolite Desacyl-Ghrelin are involved in the regulation of food intake in humans and rodents. After intraperitoneal injection, Ghrelin induces an increase in food intake as well as a significant increase in the number of c-Fos immunoreactive neurons in three hypothalamic nuclei (arcuate nucleus, dorsomedial hypothalamus, paraventricular nucleus). c-Fos immunoreactivity is a well-established marker of neuronal activation. The effect of Ghrelin on food intake is mediated via Neuropeptide Y and Agouti related peptide immunoreactive neurons. Simultaneous injection of Ghrelin and Desacyl Ghrelin, which lacks an octanoyl residue, blocks the orexinogenic effect which is exerted by injection of Ghrelin alone. Also, the number of c -Fos-immunoreactive neurons in the arcuate nucleus of the hypothalamus is significantly lower after simultaneous injection of the Ghrelin and Desacyl- Ghrelin as compared to injection of Ghrelin alone. Injection of only Desacyl- Ghrelin however does not suppress food intake. Nearly half the number of neurons activated after simultaneous injection displayed immunoreactivity for Nesfatin-1. The anorexinogenic peptide Nesfatin-1 may exert the inhibitory effect of Desacyl-Ghrelin on Ghrelin-induced food intake. The serin-threonin- kinase mammalian target of Rapamycin can be found on neurons in the arcuate nucleus. An increase in concentration of the activated, i.e. phosphorylated, form of this kinase suppresses food intake. The neuropeptide cocaine and amphetamine regulated transcript is also involved in the regulation of food intake. Like Nesfatin-1, it inhibits food intake. Double-labelling studies revealed that these peptides are intracellularly co-localized.
