Literaturverzeichnis Danksagung","Ein Nachweis von den zirkulierenden Zellen im peripheren venösen Blut kann auf eine haematogene Metastasierung hinweisen. In diesem Fall wird die Prognose als äußerst ungünstig eingestuft. Bei einem positiven Tyrosinase-mRNA-Nachweis im peripheren Blut bei Melanompatienten kann davon ausgegangen werden, dass zum Zeitpunkt der Blutentnahme eine hämatogene Dissemination von Melanomzellen stattgefunden hat. Wir untersuchten insgesamt 100 Patienten in stadium I und 46 patienten in Stadium II der Erkrankung. Die RT-PCR-Analysen ergaben bei 15 Patienten von 100 Patienten im klinischen Stadium I und bei 12 aus 46 Patienten im klinischen Stadium II den positiven Nachweis von Tyrosinase-mRNA im Blut. Im Stadium I entwickelten 3 von 15 Tyrosinase-mRNA-positiven Patienten und 2 von 85 der Tyrosinase-mRNA-negativen Patienten ein Rezidiv oder Metastasen. Im Stadium II entwickelten 2 von 12 Tyrosinase-mRNA-positiven Patienten und 8 von 34 der Tyrosinase-mRNA-negativen Patienten ein Rezidiv oder Metastasen. Im klinischen Stadium I konnte ein statistisch signifikanter Nachweis erbracht werden, dass Tyrosinase-mRNA-positiv getestete Patienten eine frühere Progression des malignen Melanoms als Tyrosinase-mRNA-negativ getestete erlitten haben. Im klinischen Stadium II konnte kein statistisch signifikanter Nachweis erbracht werden, dass Tyrosinase-mRNA-positiv getestete Patienten eine frühere Progression des malignen Melanoms als Tyrosinase-mRNA- negativ getestete erlitten haben.","Circulating tumor cells in the peripheral venous blood may be important for haematogenous spread of disease. The detection of these cells may therefore be a poor prognostic indicator. Detection of tyrosinase mRNA by RT-PCR may be of value in the early detection of dissemination. We determined the presence of tyrosinase mRNA using RT-PCR in 100 patients with stage I and 46 patients with stage II melanoma. Circulating tumor cells were detected in 15 of 100 patients in stage I and in 12 of 46 patients in stage II. In stage I were 3 of 15 patients with a positive RT-PCR-Test and 2 of 85 patients with a negative RT- PCR-Test relapses.
