In Leber- und Herzbiopsien wurden die Amyloidtypen bestimmt und die Alters- und Geschlechtsverteilung in den einzelnen Untergruppen analysiert. Außerdem wurde das Verteilungsmuster im Gewebe und der Schweregrad des Befalls bei den einzelnen Amyloidtypen ausgewertet. Im Herzkollektiv wurde zusätzlich der Anteil hereditärer ATTR-Fälle bestimmt. In einem gemischten Kollektiv wurde die Fähigkeit neuer, konformationssensitiver Substanzen zur Amyloiddetektion im Gewebe untersucht, da die Kongorot-Färbung einige Schwächen aufweist. Zum Einsatz kamen das konjugierte Polyelektrolyt polythiophene acetic acid (PTAA) und das Kamelantikörperfragment B10. Bewertet wurde der Grad der Übereinstimmung mit der Referenzfärbung Kongorot in Bezug auf die verschiedenen Organe und Amyloidtypen.","Amyloidoses are diseases characterised by deposition of misfolded proteins. The diagnosis can only be reached by pathologists and the classification is done by immunohistochemistry. We classified the type of amyloidosis in liver and endomyocardial biopsies and analysed age and gender of the patients. Furthermore we evaluated the deposition pattern and the degree of deposition. In the endomyocardial biopsies we also identified the percentage of hereditary ATTR amyloidosis. In a mixed collective we examined the ability of new conformation-sensitive agents to detect amyloid in tissue biopsies. Although Congo red still is the gold standard for amyloid detection, it has a few disadvantages. We used the conjugated polyelectrolyte polythiophene acetic acid (PTAA) and the camelid antibody fragment B10.
