Einspanier","n","2009-05-11","2018-06-07T18:40:01Z","2009-09-08T09:32:53.658Z","2009","Die Proteomik ist eine noch relativ junge Forschungsrichtung, die sich mit dem gesamten Proteinexpressionsmuster des Organismus, d.h. vor allem auch mit den Aufgaben und Wechselwirkungen der unterschiedlichsten Proteine befasst. Eine der dabei verwendeten Methoden ist die SELDI-TOF-MS (surface-enhanced-laser- deionisation/ionisation-time of-flight-mass-spectrometry). Das Anliegen der vorliegenden Arbeit ist es, im Hinblick auf eine zukünftige medizinisch- diagnostische Nutzung der SELDI-TOF-MS-Technik beim Pferd einen Einstieg in die Serum- und Liquoranalyse zu bekommen. Dazu wurde zunächst das Serum gesunder Pferde mit dem anderer Equiden und Humanserum gesunder Probanden verglichen, um einen umfassenden Eindruck vom physiologischen Peakmuster des Pferdes zu erlangen. Des Weiteren wurde Serum und Liquor systemisch und/oder neurologisch erkrankter Pferde miteinander verglichen, um erste Eindrücke der Auswirkung dieser pathologischen Zustände auf das massenspektrometrische Peakmuster des Pferdes zu bekommen. Der praktische Teil der hier vorliegenden Arbeit teilt sich in drei Teile: Im ersten Teil wurde das Peakmuster im Serum von 8 gesunden Menschen und 8 gesunden Pferden miteinander verglichen. Im zweiten Teil wurden die Spektren von jeweils 6 Pferden, Eseln und Maultieren analysiert, um eventuelle Rückschlüsse auf das verwandtschaftliche Verhältnis zu finden. Der dritte Teil bestand aus der Gegenüberstellung des Serum und Liquor von 14 neurologisch und 19 systemisch erkrankten Pferden und dem Vergleich zum Serum klinisch unauffälliger Pferde. Alle Proben wurden auf einen SAX-Chip aufgetragen und mittels SELDI-TOF-MS in einem Bereich von 4000-80000 Da untersucht. Die Peakmuster wurden miteinander verglichen und auf Zusammenhänge mit Erkrankungsbildern und anderen Parametern wie Alter, Rasse, Geschlecht hin untersucht. Zusätzlich wurden das Gesamtprotein und der Hämoglobingehalt bestimmt und von einigen Proben exemplarisch Silberfärbungen angefertigt. Serumamyloid A, Retinol-Bindungs-Protein, Transthyretin und Hämoglobin konnten zudem durch weitere Methoden der Proteinanalytik wie Western Blot oder Antikörper-Chips identifiziert werden. Im Serum der klinisch unauffälligen Pferde wurden im genannten Massenbereich 29 Peaks charakterisiert. Die Anzahl, Morphologie und Intensität der Peaks betreffend, ist beim Vergleich der Equiden, im Peakmuster der Maultiere eine deutlich größere Übereinstimmung mit den Eseln als mit den Pferden vorhanden. In der Gegenüberstellung von Serum und Liquor der zentralnervös erkrankten und neurologisch unauffälligen Pferde, weist der Liquor systemisch erkrankter Pferde weit mehr pathologische Veränderungen auf, als aufgrund der fehlenden neurologischen Symptome zu vermuten wäre. Für keinen Peak konnte bezüglich des Auftretens, der Morphologie oder der Intensität eine Korrelation zu den Parametern Geschlecht, Rasse, Alter und Erkrankung bzw. Erkrankungsdauer gefunden werden. Im Bereich 8000 bis 8500 Da tritt im Liquor zentralnervös erkrankter Pferde eine Anhäufung dicht aufeinander folgender Peaks auf, die weder im Liquor, noch im Serum anderer Gruppen beobachtet werden kann. Dies ist ein interessanter Ansatz für weitere Untersuchungen im Hinblick auf Biomarker zur Diagnostik neurologischer Erkrankungen. Die Proteindiagnostik mittels SELDI-TOF-MS scheint auch in der Veterinärmedizin eine viel versprechende Diagnostikmethode zu werden. Zum ersten Mal wurden Serum und Liquor von Pferden mit dieser Methode untersucht.","The relatively young field of research of „Proteomic“ concerns itself with the complete Protein- Expression- Patterns of the organism; here mainly with the tasks and exchanging effects of the different proteins. One of the hereby used methods is the SELDI-TOF-MS (surface-enhanced-laser-deionization/ ionization- time of-flight-mass-spectrometry). The idea of this descriptive work has been to get an introduction into the serum- and CSF- analysis for the future diagnostic use of the SELDI-TOF-MS-Technique for horses. First the serum of healthy horses has been compared to the human and to those of the equids to gain a thorough impression of the physiological peak-patterns. Further the serum and CSF of systemically and neurologically ill horses were compared to each other in order to gain first impressions of the effects of those pathological circumstances of the mass-spectrometric peak- patterns of the horse. The practical part of his work is organized in three parts: In the first part the peak- patterns in the serum of the eight healthy humans and eight healthy horses were compared to each other. In the second part the specters of six horses, six donkeys and six mules have been analyzed to draw possible conclusions concerning their relational situation. The third part consists of the confrontation of the serum and CSF of 14 neurologically ill and 19 systemically ill horses and of the comparison to the serum of clinically non-remarkable horses. All the samples have been extracted to a SAX- Chip and via the SELDI-TOF-MS examined in an area of 4000-80000 DA. The peak- patterns have been compared and examined concerning their relation of the diseases and other parameters like age, race and sex. In addition the overall protein and the hemoglobin-level were taken and some samples were colored in silver for examples. The serumamyloid A, retinol-connective- protein, transthyretin and hemoglobin could also be identified through further methods of protein- analysis like the Western Blot or Antibody-Chips. In the serum of the clinically non-remarkable horses 29 peaks in the named mass-area have been characterized. Concerning number, morphology and intensity of the peaks during the comparison among equids, the peak- patterns of the mules have shown a greater sum of common aspects with the donkeys than with the horses. In the confrontation of the serum and CSF of the horses with CNS illnesses and CNS-healthy horses, the CSF of the systemic-ill horses has shown many more pathological changes than expected due to the lack of neurological signs. A correlation to the parameters of age, race or sex concerning the appearance, morphology or intensity could not be found for any of the peaks. In the range of 8000 to 8500 Da the CSF of neurologically ill horses has shown a compiling of closely following peaks that has not been recognized in the serum and CSF of other groups. This is an interesting start for further research in the direction of bio-markers for the diagnosis of neurological diseases. The protein- diagnosis via SELDI-TOF-MS seems to become a diagnostic method for veterinary medicine as well as for humans.
