Sie kommen ubiquitär in der Umwelt vor und reichern sich aufgrund ihrer langen Halbwertzeit in der Nahrungskette an. Sie können schon in äußerst niedrigen Dosen verschiedenste Effekte in Tieren und Menschen auslösen. Ein wichtiges und sensitives Ziel der TCDD-induzierten Toxizität ist der Thymus. Hier konnten unterschiedliche Wirkungen auf verschiedene Thymozytensubpopulationen, Thymusepithelzellen sowie auf Bestandteile der EZM des Thymus beschrieben werden. In dieser Arbeit wurde untersucht, welche Wirkungen TCDD in sehr niedrigen Dosen auf Bestandteile der EZM sowie auf Integrine und TGF-beta und dessen Rezeptor im Thymus von juvenilen Marmosets hat. Hierzu wurden zum Einen juvenile Marmosets einmalig mit 100 ng/kg KG TCDD s.c. behandelt und die Thymi nach 2 (Gruppe B) bzw. 4 Wochen Beobachtung (Gruppe C) immunhistochemisch untersucht. In der 2. Versuchsreihe wurden juvenile Marmosets einmalig mit 1 (Gruppe B2) bzw. 10 ng/kg KG TCDD (Gruppe C2) s.c. behandelt und die Thymi nach 4 Wochen Beobachtung mittels Immunoblotanalysen untersucht. Nach Behandlung mit 100 ng/kg KG TCDD zeigte sich nach semiquantitativer Auswertung der Immunfluoreszenz bei nahezu allen untersuchten EZM-Proteinen des Thymus ein Anstieg der Proteinkonzentration im Vergleich zur Kontrollgruppe. Das waren im Einzelnen: Kollagen Typ I: Gruppe B +32% (Signifikanz im Student-t-Test: p<0,05) und Gruppe C +15%; Kollagen Typ IV: Gruppe B +6% und Gruppe C +24%; Fibronektin Gruppe B +15% und Gruppe C +13% und Laminin Gruppe B -8% und Gruppe C +19%. Laminin in der Gruppe B als einziges Protein verglichen mit der Kontrollgruppe vermindert. In der 2. Versuchsreihe zeigte sich nach semiquantitativer Auswertung der Immunoblots erneut fast ausschließlich ein Konzentrationsanstieg der untersuchten Proteine. Auch hier bildete die einzige Ausnahme das Laminin in der Gruppe B2, welches mit 2% vermindert war. Eine Zunahme der Proteinkonzentration im Vergleich zur Kontrollgruppe zeigten im einzelnen: CD49a: Gruppe B2 +64% und Gruppe C2 +148%; CD49e: Gruppe B2 +24% und Gruppe C2 +37%; CD49f: Gruppe B2 +41% und Gruppe C2 +74%; CD29: Gruppe B2 +20% und Gruppe C2 +44%; Kollagen Typ I: Gruppe B2 +53% und Gruppe C2 +105%; Kollagen Typ IV: Gruppe B2 +72% und Gruppe C2 +363%; Fibronektin: Gruppe B2 +55% und Gruppe C2 +89%; Laminin: Gruppe B2 -2% und Gruppe C2 +24%; TGF-beta: Gruppe B2 +31% und Gruppe C2 +102%; TGF-beta-Rezeptor: Gruppe B2 +34% und Gruppe C2 +102%. Statistisch signifikante Anstiege der Proteinkonzentrationen zeigten: CD49e und Kollagen Typ IV (alle in Gruppe C2; Signifikanz im Student-t-Test: p<0,01) sowie CD49e und Fibronektin in Gruppe B2 und CD49a, Kollagen Typ I, Fibronektin und TGF-beta-Rezeptor in Gruppe C2 (Signifikanz im Student-t-Test: p<0,05). Die beschriebenen Wirkungen von TCDD auf Integrine, die EZM sowie TGF-beta, die nach Applikation dieser sehr niedrigen Dosis auftraten, zeigen einen möglichen Mechanismus über den TCDD seine immuntoxische Wirkung entfalten kann. Diese Vermehrung der EZM ist möglicherweise durch TGF-beta und andere Zytokine vermittelt und bietet ein Erklärungsmuster für die in bereits vorliegenden Arbeiten beschriebene gestörte Thymozytenreifung und die daraus resultierenden Effekte auf das Immunsystem.","Dioxins are a group of substances, which result exclusively as unwanted by- products of the chemical industry as well as various burn processes. They occur ubiquitously in the environment and enrich themselves due to their long half-life in the food chain. They can release in extremely low doses diverse effects in animals and humans. An important and sensitives goal of the TCDD induced toxicity is the thymus. Here could be described different effects on different thymocyte populations, thymus epithelium cells as well as on components of the extracellular matrix (ECM) of the thymus. In this work was examined, which effects TCDD can cause in very low doses on components of the extracellular matrix as well as on integrins and TGF beta and its receptor in the thymus of juvenile Marmosets. For this on the one hand juvenile Marmosets with 100 ng/kg bw TCDD s.c. and the thymi after 2 (group of B) or 4 weeks observation (group of C) were immune-histochemically examined. In the 2. test series juvenile Marmosets uniquely were treated with 1 (group of B2) or 10 ng/kg bw TCDD (group of C2) s.c. and the thymi after 4 weeks observation examined by immunoblotting. After treatment with 100 ng/kg bw TCDD a rise of the protein concentration in the thymus in comparison to the control group after semi-quantitative evaluation of the immunofluorescence in almost all examined ECM proteins was notable. That were in detail: collagen type I: group B +32% (significance in the student t test: p<0,05) and group C +15%; collagen type IV: group B +6% and group C +24%; fibronectin: group B +15% and group C +13% and laminin: group B -8% and group C +19%. Laminin in the group B was the only protein with a decreased concentration compared with the control group. In the 2. test series after semi-quantitative evaluation of the immunoblots again almost all examined proteins showed an increased concentration. The only exception here also formed the laminin in the group B2, which was decreased with -2%. An increase of the protein concentration in the comparison to the control group showed in detail: CD49a: group B2 +64% and group C2 +148%; CD49e: group B2 +24% and group C2 +37%; CD49f: group B2 +41% and group C2 +74%; CD29: group of B2 +20% and group C2 +44%; collagen type I: group B2 +53% and group C2 +105%; collagen type IV: group B2 +72% and group C2 +363%; fibronectin: group B2 +55% and group C2 +89%; laminin: group B2 -2% and group C2 +24%; TGF beta: group B2 +31% and group C2 +102%; TGF beta receptor: group B2 +34% and group C2 +102%. Statistic significant rises of the protein concentrations showed: CD49e and collagen type IV (all group C2; significance in the student t test: p<0,01) as well as CD49e and fibronectin in group B2 and CD49a, collagen type I, fibronectin and TGF beta receptor in group C2 (significance in the student t test: p<0,05). The described effects of TCDD on ECM proteins, integrins and TGF beta, which arose after application of this very low dose of TCDD, is a possible mechanism of the TCDD induced immune- toxic effects.
