In den oberflächlichen Schichten des Neokortex kommunizieren Interneurone und Pyramidalzellen in einer zielzellspezifischen Weise miteinander. Die genaue Rolle der Schicht 1 Interneurone ist weiterhin Gegenstand heutiger Forschung. Nachfolgende Studien werden neben den kolumnaren Afferenzen auch die kortikokortikalen und subkortikalen Eingänge der Schicht 1 Interneurone untersuchen müssen. Insbesondere neurogliaforme Zellen scheinen im Sinne einer negativen Rückkopplung Informationen aus anderen kortikalen Arealen zu erhalten. Das Subikulum prozessiert hippokampale Information und leitet diese in verschiedene kortikale und subkortikale Areale. Zwei Typen von Projektionsneurons sind in diesem Kontext in gänzlich verschiedene Mikroschaltkreise eingebunden. Die synaptischen Eingänge aus der Area CA1 werden zielzell-spezifisch von metabotropen Glutamatrezeptoren reguliert. Nach hochfrequenter Stimulation der CA1 Afferenzen zeigen sich in beiden Pyramidalzellen ebenfalls unterschiedliche Formen synaptischer Plastizität, die nahelegen, dass hippokampale Information auf unterschiedliche Art und Weise prozessiert und in unterschiedliche Hirnregionen weitergeleitet wird. In der Area CA1 des Hippokampus wurde die Rolle verschiedener Isoformen des Proteins 4.1 untersucht. Die Ergebnisse bestätigen frühere Studien, dass das Protein 4.1 mit AMPA-Rezeptoren interagiert und diese rekrutiert. Allerdings weisen die verschiedenen Isoformen eine gewisse Redundanz auf und eine verminderte Expression einer Isoform scheint durch eine andere kompensiert werden zu können. Ein neuer Mechanismus konnte im Rahmen der homöostatischen Plastizität beschrieben werden. Es wurde gezeigt, dass ein veränderter Aufbau der AMPA-Rezeptor Heteromere in CA1 Pyramidenzellen nach langanhaltender chronischer Netzwerkinaktivierung die synaptische Übertragung und die daraus resultierende Kurzzeitplastizität maßgeblich beeinflussen kann.","The work presented includes studies on neural connectivity, synaptic transmission and plasticity in the hippocampus and neocortex. The neocortex is a six-layered structure functionally organized into columns. Understanding the so-called wiring diagram of a cortical column has received much attention in recent years. However, the role of the most superficial interneurons in layer 1 still remains unclear. We could demonstrate that neural connectivity between interneurons and pyramidal cells in the superficial layers show a high specificity of excitatory and inhibitory connections in a target cell-specific manner. We speculate that in particular neurogliaform cells may provide a powerful feed-forward inhibition of long-range intra-cortical connections. The subiculum is the main hippocampal output structure routing hippocampal information to different cortical and subcortical areas. In this context two types of projection neurons are involved in different microcircuits. The synaptic inputs from area CA1 are target cell-specifically regulated by metabotropic glutamate receptors. After high-frequency stimulation of CA1 afferents synaptic plasticity differs in these two types of pyramidal neurons, likewise suggesting that hippocampal information is processed in different ways and possibly forwarded in different brain regions. In area CA1 of the hippocampus we examined the role of different isoforms of protein 4.1. The results confirm previous studies demonstrating the interaction of protein 4.1 with AMPA receptors. However, the different isoforms exhibit a certain redundancy and the reduced expression of an isoform seems to be compensated by another isoform. A new homeostatic mechanism is described following long- lasting inactivation of the network activity.
