Dies führt histologisch zu degenerativen Veränderungen der Skelettmuskulatur, die neben Fasernekrosen-und Regeneraten eine progrediente endo- und perimysiale Fibrose zur Folge haben. Bei der Zunahme der extrazellulären Matrix (EZM) steht die Akkumulation der fibrillären Kollagene I-III im Vordergrund. Diese Organfibrose kann einerseits durch eine überproportionale Ablagerung von Kollagenen verursacht werden, andererseits kann auch ein gestörter Matrixabbau (Fibrolyse) zu einer Akkumulation von fibrösem Gewebe führen. Die zinkabhängigen Metalloproteinasen haben ein breites Substratspektrum und sind gemeinsam in der Lage, die gesamten Komponenten der extrazellulären Matrix zu hydrolisieren, wobei hauptsächlich die Kollagenasen für den Abbau der fibrillären Kollagene (I-III) verantwortlich sind. Die Aktivität der MMP wird durch deren spezifischen Inhibitoren, den „tissue inhibitors of matrix-metalloproteinases“ gehemmt. Gegenstand der vorliegenden Arbeit ist die Analyse der Proteinexpression von MMP-1, -2 und den Inhibitoren TIMP-1 und -2 im DMD-Muskel mittels Immunhistochemie und Western-Blot. Die Expressionsmuster sprechen dafür, dass MMP-1 u. a. durch vermehrt gebildetes TIMP-1, TIMP-2 und LIMP gehemmt wird und dadurch der vermehrten Synthese von Kollagen Typ I und III nicht suffizient entgegen gewirkt werden kann. Es erscheint daher sinnvoll, antifibrotisch wirksame Substanzen in die therapeutischen Überlegungen mit einzubeziehen.","Duchenne Muscular Dystrophy (DMD) is caused by mutations in the DMD gene that leads to absence of a functional dystrophin protein at the muscle cell membrane and to degenerative histological changes of skeletal muscle, which, in addition to necrotic and regenerating fibers has a progressive endo-and perimysial fibrosis to follow. The present work is the analysis of protein expression of MMP-1, -2 and the inhibitors TIMP-1 and -2 in DMD muscle by immunohistochemistry and Western blot. The expression patterns suggest that MMP-1 is inhibited by increased TIMP-1, TIMP-2 and LIMP and therefore can not sufficiently counteract the increased synthesis of type I and III collagen.
