Um entzündungsspezifische Änderungen der AKO- Inhaltsstoffe zu erfassen, sollten gezielt Atemkondensatproben von Kälber mit natürlich erworbenen (Chlamydien-assoziierten) und experimentell induzierten (Mycoplasma bovis) respiratorischen Affektionen untersucht und mit AKO-Proben von entsprechenden Kontrolltieren verglichen werden. Die Gewinnung von AKO- Proben erfolgte an stehenden unsedierten Kälbern, deren exspiratorischer Atemstrom mit Hilfe einer Maske in eine Kühlfalle in Form des kommerziell verfügbaren Atemkondensat-Sammelgerätes ECoScreen (Viasys Healthcare, Höchberg) geleitet wurde. Parallel zu jeder AKO-Gewinnung wurden mittels eines im Gerätesystem integrierten elektronischen Spirometers ( ECoVent ) folgende Kenngrößen der Spontanatmung erfasst: die maximale exspiratorische Atmungsstromstärke [l/s], das exspiratorische Atemzugvolumen [l], das exspiratorische Atemminutenvolumen [l], die Atmungsfrequenz [Atemzüge/min]. In der BALF von Kälbern mit Chlamydia-psittaci-Infektion waren signifikant höhere Konzentrationen an Gesamtprotein, 8-Isoprostan und Harnstoff sowie eine niedrigere Nitrit-Konzentration nachweisbar. Diese Befunde sprechen für entzündlich bedingte Veränderungen im epithelialen Oberflächenfilm der Atemwege. Im AKO konnten weder für die Konzentrationen noch für die pro 100 l exhalierten Mengen der identischen Inhaltsstoffe signifikante Unterschiede zwischen den Tiergruppen nachgewiesen werden. Das exhalierte Ammonium hingegen war im AKO der Chlamydia-positiven Kälber signifikant geringer als im AKO der Chlamydia-negativen Tiere. Eine post infectionem erhöhte Abscheidung des Leukotrien B4 je 100 Liter Exhalat und der verminderte pH-Wert bei den experimentell mit Mycoplasma bovis infizierten Tieren weisen auf einen akuten Entzündungsvorgang innerhalb der Atemwege hin. Atemkondensat, BALF und Blut unterscheiden sich bezüglich ihres biologischen und diagnostischen Informationsgehaltes. Entzündungsvorgängen innerhalb der Lunge wurden durch die BALF sensitiver widergespiegelt als durch AKO. Mögliche Ursachen hierfür könnten sowohl Einflüsse des Atmungsmusters als auch bisher nicht zu standardisierende Verdünnungseffekte auf die Zusammensetzung des AKO sein. Weiterführende Untersuchungen sind notwendig, um die Gewinnung des Atemkondensates zu standardisieren und die Eignung der Inhaltsstoffe zu prüfen, entzündliche Vorgänge auf den Atemwegen darzustellen.","Evaluation of different components of the exhaled breath condensate and their potential to reflect pulmonary inflammations in calves The present study aimed to examine the following exhaled breath condensate (EBC) components in calves: total protein, 8-isoprostane, leukotriene B4, nitrite, urea, ammonia, concentration of protons (pH). In order to evaluate their potential to reflect inflammatory pulmonary processes, three consecutive steps were defined: After the knowledge of methodological and biological background of EBC collection, two infection models were used to clarify which components of EBC might be able to indicate pulmonary inflammation. Thus, calves naturally infected with Chlamydiaceae as well as calves experimentally infected with Mycoplasma bovis were compared to healthy controls. EBC samples were collected in conscious animals during spontaneous breathing using a face mask and the commercially available EBC collection system ECoScreen (Viasys Healthcare, Hoechberg, Germany). During each EBC collection, an integrated electronic spirometer ( ECoVent ) was explored to measure the following parameters of the spontaneous ventilation: maximal expiratory peakflow [l/s], expiratory tidal volume [l], expiratory minute volume [l], and respiratory rate [breath/min]. In the group of Chlamydia-psittaci-positive calves, significantly higher concentrations of total protein, 8-isoprostan and urea but a lower concentration of nitrite was measured in BALF samples in comparison to control calves without chlamydial infection. These results may indicate changes in the airway epithelial lining fluid caused by inflammatory processes. With respect to EBC, only the exhaled ammonia was significantly lower in EBC samples of Chlamydia-positive calves. For all other EBC components, neither concentrations expressed per ml EBC nor those related to 100 l exhaled breath differed significantly between the two animal groups. The experimentally induced M. bovis infection led to elevated concentrations of leukotriene B4 per 100 litres exhaled breath and to lower pH-values indicating inflammatory processes inn the lung that were not detectable by clinical symptoms. The diagnostic samples EBC, BALF and blood serum differ with respect to their diagnostic information. BALF was found to be more reliable in reflecting inflammatory processes within the lung. For EBC, one can assume that unsolved questions of ventilatory influences or dilution effects on the composition of EBC interfere with diagnostic information.
