Die Lokalisation und Bedeutung von Leukozyten in der Cochlea während der Entwicklung sind bisher noch nicht genau bekannt. Das Ziel der vorliegenden Arbeit war es, Leukozyten während der postnatalen Reifung der Cochlea im Rattenmodell zu untersuchen und einen möglichen Zusammenhang mit Cisplatin-induzierter Ototoxizität herzustellen. Methodik Cochleae von Wistar-Ratten des 1., 3., 5., 7. und 9. postnatalen Tages wurden zur Herstellung organotypischer Explantatkulturen isoliert. Relevante Immunzellen wurden an Kryoschnitten mittels Antikörpernachweis der CD45-, CD11b-, CD68- und CD117/c- Kit-Expression und Avidin-Färbung von intrazellulärer Heparingranula in den Mastzellen nachgewiesen und ausgezählt. Die Cochlea-Kulturen wurden mit 20 μM Cisplatin inkubiert, um den Effekt auf die Immunzellpopulationen zu untersuchen. Ergebnisse CD45-positive, CD11b-positive, CD68-positive und CD117/c-Kit-positive Zellen wurden in der Cochlea nachgewiesen. Die Leukozyten waren in unterschiedlichen Abschnitten der Cochlea lokalisiert: CD68+- und CD11b+-Zellen befanden sich überwiegend im Limbus spirale und im Modiolus, CD117/c-Kit+- und CD45+-Zellen überwiegend im Modiolus. Die Anzahl der Leukozyten veränderte sich während der postnatalen Entwicklung. CD45+-Zellen nahmen im apikalen Teil bis Tag 5 zu und blieben danach konstant, während sie im medialen Teil ab Tag 5 abnahmen. CD11b+- Zellen nahmen beginnend mit pT1 stetig ab und waren nach neun Tagen nur noch im Modiolus und im Ligamentum spirale nachweisbar. CD68+-Zellen nahmen während der ersten drei Tage zu, danach stetig ab. Die Inkubation der Cochlea-Explantate mit 20 μM Cisplatin resultierte in einer Schädigung der Mastzellen, der CD45+-, CD11b+- und CD68+-Zellen und der Haarzellen. Schlussfolgerung Die Ergebnisse der vorliegenden Arbeit demonstrieren die Existenz residenter Leukozyten in der Cochlea und eine Veränderung der Zellzahlen während der postnatalen Entwicklung. Die Schädigung dieser Zellen durch Cisplatin könnte auf einen Zusammenhang mit Cisplatin-induzierter Ototoxizität hindeuten.","Introduction The inner ear contains various immune cells that may contribute to immune defense and development but also mediate pathological degeneration. The localization and importance of leukocytes in the cochlea during development have not been well understood. The aim of the present work was to investigate leukocytes during postnatal maturation of the cochlea in a rat model and to establish a possible link with cisplatininduced ototoxicity. Methodology Cochleae from Wistar rats of postnatal days 1, 3, 5, 7, and 9 were isolated to generate organotypic explant cultures. Relevant immune cells were detected using antibodies against CD45, CD11b, CD68, or CD117/c kit, as well as avidin staining. The positive cells were scored in cochlear cryosections. Cochlear cultures were exposed to 20 μM cisplatin to examine the effect on immune cell populations. Results The resident CD45-positive, CD11b-positive, CD68-positive, and CD117/c-Kit positive cells were identified in different cochlear sections. CD68+ and CD11b+ cells were found predominantly in the spiral limbus and modiolus, while CD117/c-Kit+ and CD45+ cells were primarily located in the modiolus. The number of leukocytes changed during postnatal development. In detail, the number of CD45+ cells in the apical part increased until postnatal day five and remained constant after that. In contrast, the number of CD45+ cells in the medial part of the cochlea decreased after the fifth postnatal day. The number of CD11b+ cells steadily decreased from the first day after birth, and in p9 animals, the CD11b+ cells were detectable only in the modiolus and spiral ligament. The number of CD68+ cells increased during the first three days and steadily decreased after that. Incubation of cochlear explants with 20 μM cisplatin resulted in damage to mast cells, CD45+ cells, CD11b+ cells, CD68+ cells, and hair cells. Conclusion The present work results demonstrate the existence of resident leukocytes in the cochlea and a change in cell numbers during postnatal development.
