Dieser Umstand hat dazu geführt, dass trotz der unmittelbaren Bedrohung durch den Bioterrorismus bis zum heutigen Tage nur wenige zuverlässige diagnostische Mittel zur Detektion von nativen Bioterrorstoffen verfügbar sind. Im Zuge der Arbeit wurden durch die rekombinante DNA- Technologie synthetische Vektoren geschaffen, die eine Expression von nativen Proteinkonformationen für zwei Kandidatenagenzien, BoNT und die Glykoproteine (GP) der Filoviren MarV und EboV, ermöglichten. Die synthetischen Nukleinsäuresequenzen wurden hierbei für den Einsatz in Säugern kodonoptimiert und für eine optimale Verwendung durch Mutation der Ursprungssequenz modifiziert. Für BoNT A1 wurden zwei Modifikationen der Aminosäureabfolge vorgenommen, um durch die Inhibition der Zink-Bindung eine vollständige Entgiftung des Enzyms zu erreichen. Die filoviralen GP-Konstrukte wurden durch Entfernung einer slipperysite (EboV) auf die Expression des Volllängenglykoproteins Gp0 optimiert. Nach dem Aufbau und der Evaluierung von nativen Selektionssystemen, durch einen capture Assay, unter Verwendung eines GP-Fc-Fusionsproteins, wurden die so erzeugten Konstrukte erfolgreich zur Immunisierung und Induktion von mono-/ polyklonalen Antikörpern eingesetzt. Vier monoklonale Antikörper des Isotyps IgG gegen BoNT A1 und vier monoklonale Antikörper des Isotyps IgM gegen EboV Gp wurden in Bezug auf Detektionseigenschaften feincharakterisiert und z.T. zur Bildung von 14 autonomen Detektionspaaren eingesetzt. Das Repertoire der generierten Antikörper umfasste Antikörper mit Spezifität gegen die leichte und schwere Kette des BoNT A1, das Gp1 und Gp2 des EboV Gp, sowie zwei Antikörper mit Konformationsabhänigkeit der Epitope. Die Testung der generierten monoklonalen Reagenzien gegen BoNT zeigte eine effiziente Erkennung bis zu Nachweisgrenzen im Bereich von 8,5 - 25 pg BoNT A1/ ml und eine hochaffine Bindung nativen BT- Antigens. Die Überprüfung der Kreuzreaktivität ermittelte eine spezifische Detektion von nativen hochtoxischen BoNT-Komplexen der Toxinsubtypen A1-A3, aus clostridialen Kulturüberständen, durch drei der vier monoklonalen Detektionsantikörper. Zusätzlich konnten erfolgreich Versuchsreihen durchgeführt werden, um neuartige genetische Zytokinadjuvantien im aviären System zu testen und zu etablieren, was zu einer effizienten Steigerung der spezifischen IgY-Titer und der Gewinnung von reaktiven IgY- Präparationen im Gramm-Bereich führte. Ein im Zuge der Arbeit entwickelter Selektionstest für filovirusreaktive Antikörper zeigte verlässliche Eigenschaften im Einsatz als diagnostischer Antikörpersuchtest. Das Verfahren bietet eine Grundlage für die Entwicklung neuartiger antikörperbasierter klinischer Filovirus- Nachweissysteme.","The unifying characteristic of bioterroristic agents is their capacity to cause severe injury to health upon infection or ingestion. Despite the threat of bioterrorist attacks, only a few reliable diagnostic systems for the detection of native bioterror agents are to date commercially available. During the course of this project, synthetic expression vectors were created by recombinant DNA-technology that permit the expression of proteins with native conformation from two candidate agents: BoNT A1 and the glycoproteins of the Filoviridae MarV & EboV. The synthetic nucleic acid sequences were engineered for optimal expression through mammalian codon-optimization and minor mutations. In addition, the BoNT A1 was rendered completely non-toxic by the introduction of two codon modifications that inhibit the enzyme's zinc binding capability. Filoviral Gp- constructs were optimized for expression of the full-length glycoprotein Gp0 through elimination of a slippery site (EboV). Following production and evaluation of a native screening system by a capture assay employing a Gp-fc fusion protein, the vector DNA was used to immunize laboratory animals to facilitate the preparation of monoclonal and polyclonal antibodies. Four IgG monoclonal antibodies specific for BoNT A1 and four IgM specific for EboV Gp were characterized in detail and used to set up 14 autonomous detection pairs. The antibodies generated included some recognizing the light and heavy chains of BoNT A1, others recognizing the Gp1 and Gp2 of the EboV Gp and two antibodies suspected to bind conformational epitopes. The monoclonal reagents specific for BoNT A1 could detect as little as 8.5 – 25 pg BoNT A1/ml and had a high affinity for the native BT-antigen. An analysis of cross-reactivity revealed a specific detection of highly toxic native BoNT complexes of the toxin subtypes A1-A3 from clostridial culture supernatants by three of the four monoclonal antibodies. Additionally, a series of animal trials was performed to evaluate and establish novel genetic cytokine adjuvants in the avian system that resulted in a significant enhancement of specific IgY titers and facilitated the production of IgY in high quantities. During the course of this work, a filovirus-reactive antibody screening assay was developed and was shown to be versatile and reliable for application as a diagnostic antibody screening test.
