Hübner","n","2008-10-14","2018-06-07T17:05:49Z","2009-04-16T12:33:10.936Z","2009","In dieser Arbeit wurden nach computerbasierter Analyse alle putativen, evolutionär konservierten Bindungsstellen des humanen und bovinen ECE-1a Promotors für den Transkriptionsfaktor Ets-1 bzw. für basal exprimierte ETS- Transkriptionsfaktoren durch das Gelshiftverfahren (EMSA) untersucht. Dabei konnte für die humanen Konsenssequenzen -226, -639, -659, -787, -816, -972 und für die bovinen Konsenssequenzen -602 und -622 eine Bindung eines basal exprimierten ETS-Transkriptionsfaktors gezeigt werden. Um das Verhalten des ECE-1a-Promotors zu untersuchen, wurden die nativen Bindungssequenzen in Transfektionsstudien nach Einbringung von Mutationen funktionell analysiert. Gemeinsam mit den Ergebnissen der Protein-DNA-Interaktionsstudien mittels EMSA konnte die Wirkung der Mutation der jeweiligen Bindungsstellen funktionell charakterisiert werden. Die Ergebnisse deuten darauf hin, dass die Expression der humanen Isoform ECE-1a in der Endothelzelllinie EA.hy926 unter basalen Bedingungen durch die Bindung eines Repressors an den Promotor reguliert wird. Die bovine ECE-1a Isoform wird hingegen nicht durch einen Repressor reguliert, wie Transfektionsstudien an bovinen aortalen Endothelzellen (BAEC) zeigen. Weiterhin konnte gezeigt werden, dass zur Vermittlung der inhibitorischen Wirkung des Repressors Wechselwirkungen zwischen der im 5’-Bereich des Promotors gelegenen Region (-1086 bis -964) und anderen Regionen des Promotors notwendig sind (-787, -226). Dies deutet darauf hin, dass die Wirkung des Repressors unter Bildung von Komplexen mit weiteren Proteinen stattfindet. Zur Identifizierung des Repressors und dem zugrunde liegenden Mechanismus der Inhibition wurden zwei verschiedene Modelle diskutiert. Es handelt sich unter Einbeziehung der bekannten Literatur entweder um eine unbekannte Spleißvariante von Ets-1 oder ein anderes ETS-Protein, möglicherweise NERF-1. Die vorliegende Arbeit erweitert das Verständnis der isoformspezifischen Regulierung von Promotoren. Es wurden Unterschiede in Bezug auf die humane und bovine Spezies dargestellt. Ein weiterer Schritt wäre die Identifikation des bisher unbekannten, postulierten Repressors, um das Verständnis der direkten Inhibition von ETS-Transkriptionsfaktoren zu erweitern.","In the work presented here, all putative, evolutionarily conserved binding sites of the human and bovine ECE 1a promoter were investigated for the transcription factor Ets-1 or for basally expressed ETS transcription factors using the gel shift method (EMSA) following a computer-based analysis. A binding was shown of a basally expressed ETS transcription factor for the human consensus sequences -226, -639, -659, -787, -816, -972 and the bovine consensus sequences -602 and -622. To investigate the behaviour of the ECE 1a promoter, a functional analysis was carried out of the native binding sequences in the transfection study after the introduction of mutations. Together with the results of the protein DNA interaction studies using EMSA, the action of the mutation of each binding site was functionally characterised. The results indicate that, under basal conditions, the expression of the human isoform ECE-1a in the endothelial cell line EA.hy926 is regulated by the binding of a repressor to the promoter. In contrast, the bovine ECE 1a isoform is not regulated by a repressor, as shown in transfection studies on bovine aortal endothelial cells (BAECs). Furthermore, it was shown that to promote the inhibitory action of the repressor, interactions are necessary between the 5’ region (-1086 to -964) and other regions of the promoter (-787, -226). This indicates that the effect of the repressor occurs in conjunction with the formation of complexes with other proteins. Two different models were discussed for the identification of the repressor and the underlying inhibition mechanism. With reference to known literature, either an unknown spliced variant of the Ets-1 or another ETS protein, possibly NERF-1, is involved. The present work expands the understanding of the isoform-specific regulation of promoters. Differences were shown with respect to the human and bovine species.
