Häufiger sind sie jedoch mit der Neurofibromatose vom Typ 1 (NF1) assoziiert. Die Prognose von MPNST ist unverändert ungünstig, da die Tumoren zumeist chirurgisch nicht resezierbar sind. Eine adjuvante Therapie mit Chemo- und Radiotherapie kann das Überleben kaum beeinflussen. Für MPNST gibt es zur Zeit kein Xenotransplantatmodell. Der Heparin-bindende Wachstumsfaktor Midkine (MK) scheint wesentlich an der Tumorgenese von Neurofibromen und MPNST beteiligt zu sein. MK wirkt mitogen, anti-apoptotisch, angiogen und fördert die Tumorigenität von Zelllinien neuroektodermalen Ursprungs. Zielsetzung dieser Arbeit war es, den Einfluss von MK auf das maligne Verhalten und besonders die Tumorigenität von MPNST zu untersuchen. Hierzu wurden MPNST-Zelllinien stabil transfiziert. Zum Nachweis der Überexpression von MK auf der Ebene der Ribonukleinsäuren (RNS) wurde eine reverse Transkription mit anschließender quantitativer Polymerasenkettenreaktion (quantitative RT-PCR) etabliert. Zur Messung der Proteinkonzentration von MK im konditionierten Medium wurde ein Enzym-Immunoassay (ELISA) verwendet. Die Analyse der Wachstumsraten der verschiedenen Zelllinien erfolgte mit einem Dimethylthiazol- carboxymethoxyphenyl-sulfophenyl-tetrazolium-Assay (MTS-Assay). Die Chemosensitivität der Zelllinien gegenüber Vincristin wurde ebenfalls mit dieser Methode untersucht. Zur Apoptosemessung wurde ein TUNEL- ( terminal deoxynucleotidyltransferase dUTP nick end labeling -) Assay etabliert, da die Messung durch Färbung mit fluoreszenzmarkierten Annexin und Propidiumjodid keine eindeutigen Interpretationen ermöglichte. MPNST-Zelllinien sind nicht tumorigen. Auch die stabile Überexpression von MK in S462- sowie in ST88-14-Zellen war nicht ausreichend, um ein Tumorwachstum in Nod/Scid-( non- obese diabetes/severe combined immunodeficiency -) Mäusen zu induzieren. Dafür erhöhte die Überexpression von MK in der MPNST Zelllinie S462 die Zellvitalität unter Serumentzug. Auch externe Addition von MK steigerte die Vitalität der MPNST-Zellen. Die Überexpression von MK schützte S462-Zellen vor durch Serumentzug induzierter Apoptose und ist wahrscheinlich als Ursache für die erhöhte Vitalität anzusehen. Eine Stimulation des Wachstums wurde durch MK nicht induziert. Außerdem führte die Transfektion von MK zu einer Abnahme der Chemosensitivität gegenüber Vincristin. Konditioniertes Medium von mit MK transfizierten S462-Zellen war ein potentes Mitogen für humane venöse Endothelzellen (HUVEC). Auch andere angiogene Faktoren im konditionierten Medium könnten neben MK für die Stimulierung des Wachstums der HUVEC verantwortlich sein. So war die MK-Überexpression in S462-Zellen begleitet von höheren RNS-Konzentrationen des vaskulären endothelialen Wachstumsfaktors (VEGF). Zusammenfassend zeigen diese Ergebnisse, dass die stabile Transfektion von MK in S462-Zellen das Überleben steigert und die angiogene Potenz erhöht. Die Entwicklung spezifischer Inhibitoren von MK als Teil eines neuen therapeutischen Ansatzes zur Behandlung von MPNST ist daher möglicherweise vielversprechend.","Malignant peripheral nerve sheath tumors (MPNST) are aggressive soft tissue tumors arising sporadically, but more frequently in patients with Neurofibromatosis type 1 (NF1). Prognosis of MPNST is dismal because most of the tumors are surgically not resectable. So far adjuvant therapy using chemotherapy and radiation has only little influence on the survival rate of MPNST patients. The heparin-binding growth factor Midkine (MK) is implicated in the tumorigenesis of neurofibromas and MPNST. MK is mitogenic, anti- apoptotic, angiogenic and promotes tumorigenicity in tumor cells of neuroectodermal origin. So far, there is no xenograft animal model available. Thus, we investigated the role of MK in terms of tumorigenicity and malignant biology in MPNST cells using stable transfection of MK into MPNST cell lines. For the detection of MK overexpression on the ribonucleic acid (RNA) level a reverse transcription with following quantitative polymerase chain reaction (quantitative RT-PCR) was etablished. The concentration of MK protein in conditioned medium was determinened using an enzyme-linked immunosorbent assay (ELISA) normally used to determine MK in patient samples. The growth rate of MPNST cells and human umbilical venous endothelial cells (HUVEC) was analysed using a dimethylthiazol-carboxymethoxyphenyl-sulfophenyl-tetrazolium (MTS) assay. Also, the chemosensitivity to vincristine of MPNST cells was analysed using this method. A terminal deoxynucleotidyltransferase dUTP nick end labeling (TUNEL) assay was established for the detection of apoptosis, as fluorescence tagged annexin and propidium iodide yielded conflicting results. Current MPNST cell lines are not tumorigenic. Stable overexpression of MK into the cell lines S462 and ST88-14 was insufficient to induce tumor growth in a non-obese diabetes/severe combined immunodeficiency (nod/scid) mouse xenotransplant model. But, overexpression of MK in the MPNST cell line S462 increased cell viability when stressed by serum deprivation. Also, the external addition of MK increased the viability of the cells. Stable transfection of MK protected S462 cells of apoptosis induced by serum deprivation and probably caused the increased viability. MK overexpression could not increase the growth rate in transfected cells. Additionally, the chemosensitity to vincristine was decreased after stable transfection of MK. Conditioned medium of MK overexpressing S462 cells was a potent mitogen for human venous endothelial cells (HUVEC). Furthermore, MK overexpression in S462 cells was accompanied by higher levels of vascular endothelial growth factor (VEGF) RNA suggesting that other angiogenic factors induced by MK overexpression participated in the stimulation of HUVEC growth rate. These results demonstrate that stable transfection of MK in S462 MPNST cells increases their survival and angiogenic potency.
