Eine Störung des Redoxgleichgewichts kann zu Krankheiten wie Arteriosklerose oder Krebs führen. Die Glutathionperoxidase-4 (Gpx4) und die 15-Lipoxygenase (Alox15) sind funktionelle Gegenspieler im Redoxsystem tierischer Zellen. Die Gpx4 reduziert komplexe Hydroperoxylipide, wirkt als endogener Alox15-Inhibitor und fungiert gleichzeitig als Strukturprotein. Systemische Inaktivierung des Gpx4-Gens führt zur Letalität homozygoter Knockout-Embryonen, während sich Alox15 -Knockout-Mäuse normal entwickeln. Ein spermienspezifischer Knockout des Gpx4-Gens führt zur Infertilität männlicher Mäuse. Bislang war unklar, ob die embryonale Letalität der Gpx4-defizienten Mäuse auf den Mangel an Gpx4-Aktivität oder auf das Fehlen des Strukturproteins zurückzuführen ist. Es konnte nicht geklärt werden, ob Gpx4 und Alox15 auch in vivo als funktionelle Gegenspieler wirken und ob ein funktioneller Knockout der Alox15 die Defizite der Inaktivierung des Gpx4-Gens rückgängig machen kann. Um diese Fragen zu beantworten, haben wir ein murines Knockin-Modell hergestellt, bei dem das Gpx4-Gen durch ein Konstrukt ersetzt wurde, das für ein katalytisch inaktives Enzym (U46A Gpx4) kodiert. Mit diesem Modell wurden die embryonale Letalität homozygoter Embryonen und die Fertilität heterozygoter Merkmalsträger untersucht. Um den Effekt der Inaktivierung der Alox15 auf diese Knockin-Mäuse zu untersuchen, wurde in einem zweiten Schritt ein homozygoter Alox15-Knockout in die Gpx4 defizienten Mäuse eingekreuzt. Heterozygote Gpx4-Knockin-Mäuse (U46A Gpx4-/++Alox15+/+) entwickeln sich normal, während homozygote U46A Gpx4+/++Alox15+/+-Mäuse zwischen Embryonaltag 6.5 und 7.5 absterben. Die embryonale Letalität konnte durch einen zusätzlichen Knockout der Alox15 (U46A Gpx4-/++Alox15-/-) nicht kompensiert werden. Männliche U46A Gpx4-/++Alox15+/+-Mäuse weisen eine reduzierte Fertilität auf, die sich durch eine verminderte Motilität ihrer Spermien und durch morphologische Veränderungen in den Mittelstücken manifestieren. Die reduzierte Fertilität der U46A Gpx4-/++Alox15+/+-Mäuse und die schlechtere Motilität ihrer Spermien konnte durch einen zusätzlichen homozygoten Knockout der Alox15 (U46A Gpx4-/++Alox15-/-) ausgeglichen werden. Bei einzelnen Individuen kam es zudem zur Normalisierung der morphologischen Veränderungen. Diese Ergebnisse zeigen, dass die embryonale Letalität Gpx4-defizienter Maus-Embryonen durch den Verlust der katalytischen Aktivität der Gpx4 (U46A Gpx4+/++Alox15+/+) verursacht wird. Die embryonale Letalität konnte durch einen zusätzlichen homozygoten Knockout der Alox15 (U46A Gpx4+/++Alox15-/-) nicht kompensiert werden. Jedoch konnte die Subfertilität heterozygoter Gpx4-Knockin-Männchen (U46A Gpx4-/++Alox15+/+) und die verminderte Motilität ihrer Spermien durch einen zusätzlichen homozygoten Knockout der Alox15 (U46A Gpx4-/++Alox15-/-) ausgeglichen werden.","The redox-status is important for the expression pattern of cells and thus it contributes to the regulation of the cellular metabolism. Dysregulation in the cellular redox homeostasis has been related to the pathogenesis of various diseases such as atherosclerosis and cancer. Alox15 and Gpx4 are functional counterplayers in metabolism of complex hydroperoxy lipids. Gpx4 reduces Alox15-derived hydroperoxy lipids, acts as endogenous Alox15 inhibitor but also functions as structural protein (moonlighting character). Homozygous Gpx4-deficient animals die at an early stages of embryogenesis and sperm specific disruption of the Gpx4 gene leads to male infertility. Because of the moonlighting character of Gpx4 it remained unclear whether the lack of catalytic Gpx4 activity or the deficiency of the structural protein might be considered the major cause of embryonic lethality. Moreover, it has not been explored in the past whether systemic inactivation of the Alox15 gene might reverse the defective phenotypes induced by Gpx4 deficiency. To address these questions we created Gpx4 knockin mice, in which the Gpx4 gene was replaced by a mutant construct that encodes for a catalytically silent enzyme variant (U46A Gpx4). In a second step viable heterozygous Gpx4 knockin mice (U46A Gpx4-/+) were crossbred with Alox15 knockout mice (Alox15-/-) to explore the impact of Alox15 on Gpx4 deficiency. Employing these Gpx4 knockin mice we found that homozygous animals (U46A Gpx4+/++Alox15+/+) die in utero between E6.5-7.5 and additional Alox15-deficient (U46A Gpx4-/++Alox15-/-) did not rescue embryonic lethality. In contrast, heterozygous knockin mice (U46A Gpx4-/++Alox15+/+) were viable but sperms extracted from the epididymal cauda of U46A Gpx4+/-+Alox15+/+ mice exhibited impaired motility characteristics. Detailed morphological studies including immune electron microscopy indicated structural midpiece alterations within the mitochondrial capsule. When U46A Gpx4+/--mice were crossed with Alox-deficient animals the resulting males (U46A Gpx4+/-+Alox15-/-) exhibited improved fertility and motility characteristics. In some individuals we even detected wildtype-like morphological sperm characteristics. Taken together these data indicate that Alox15 deficiency does not rescue the lethal phenotype of U46A Gpx4+/+ knockin mice. Heterozygous expression of a catalytic inactive Gpx4 variant (U46A Gpx4-/++Alox15+/+) impairs male but not female fertility.
