Sie üben sowohl zelladhäsive als auch regulatorische Funktionen aus. Alle Mitglieder der Protocadherine besitzen eine variable extrazelluläre Domäne und eine größtenteils identische zytoplasmatische Domäne. Protocadherine kommen hauptsächlich im zentralen Nervensystem vor und scheinen in den Ablauf der synaptischen Signalübertragung und Synapsenerhaltung involviert zu sein. Ziel dieser Arbeit war es, mittels des Hefe-2-Hybriden Systems einen potentiellen intrazellulären Protein-Protein- Interaktionspartner des Protocadherins γC5 zu identifizieren. Dazu wurde eine cDNA-Bank aus murinem Gehirn konstruiert und auf Interaktion mit dem konstanten, cytoplasmatischen Teil des Protocadherin γC5 getestet. Als potentieller Interaktionspartner konnte Calmodulin (Calm1) identifiziert werden, welches durch weitere Versuche analysiert und verifiziert werden muß.","Protocadherins belong to the superfamily of cadherins and can be subdivided into an alpha-, beta- and gamma cluster. They show both celladhesive as well as regulatory functions. All protocadherin members possess a variable extracellular domain and a mainly identical cytoplasmic domain. Protocadherins are preeminently expressed in the central nervous system and seem to be involved in synaptic signalling and the structural maintenance of synapses. Aim of this work was to identify a potential intracellular protein-protein- interaction partner of protocadherin gamma C5 by employing the yeast-2-hybrid system. Thus, a murine brain cDNA library was constructed and tested for positive interaction with the cytoplasmic part of protocadherin gamma c5.
