One of the problems encountered when developing AMPs as drugs is the difficulty of reaching sufficient killing concentrations under physiological conditions. Here, using pexiganan, a cationic peptide derived from a host defense peptide of the African clawed frog and the first AMP developed into an antibacterial drug, we studied whether sub-lethal effects of AMPs can be harnessed to devise treatment combinations. We studied the pexiganan stress response ofStaphylococcus aureusat sub-lethal concentrations using quantitative proteomics. Several proteins involved in nucleotide metabolism were elevated, suggesting a metabolic demand. We then show thatStaphylococcus aureusis highly susceptible to antimetabolite nucleoside analogs when exposed to pexiganan, even at sub-inhibitory concentrations. These findings could be used to enhance pexiganan potency while decreasing the risk of resistance emergence, and our findings can likely be extended to other antimicrobial peptides.","13 Seiten","https://refubium.fu-berlin.de/handle/fub188/28905||http://dx.doi.org/10.17169/refubium-28654","eng","https://creativecommons.org/licenses/by/4.0/","500 Naturwissenschaften und Mathematik::570 Biowissenschaften; Biologie::570 Biowissenschaften; Biologie","pexiganan||antibiotic resistance||antimetabolites||antimicrobial peptides||antibiotics||nuceloside analogs","Antimicrobial Peptide Induced-Stress Renders Staphylococcus aureus Susceptible to Toxic Nucleoside Analogs","Wissenschaftlicher Artikel","open access","1686","10.3389/fimmu.2020.01686","Frontiers in Immunology","https://doi.org/10.3389/fimmu.2020.01686","11","1664-3224","Biologie, Chemie, Pharmazie","Institut für Biologie:::77195c3c-c355-4e73-837e-d2e0ba8b6c51:::600","no","WoS-Alert"
