Summary 111 Bibliography 115 Danksagung","The emerging field of plasmonics that treats the interaction of light with oscillating electrons in a metal surface or particle is a hot topic in surface science. The confinement of (visible) light to nanometer regions by the excitation of localized surface plasmons opens up exciting research routes as it overcomes the Abbe optical resolution limit of $lambda$/2. Based on plasmonics, tip-enhanced Raman spectroscopy (TERS) meets one of the long- expected goals in surface science: The simultaneous topographic and chemical investigation of a nanometer-sized sample region is rendered possible by the combination of scanning probe microscopy and Raman spectroscopy. (Sub)monolayer adsorbates at atomically smooth surfaces can be investigated effortlessly, even down to the single-molecule level. In this thesis, several approaches are followed to obtain a deeper insight into the physical, chemical and technical aspects of TERS. In order to gain understanding of the near- field distribution in the tip-substrate cavity, tip-sample distance dependence experiments are carried out. TER spectra are analyzed regarding the behaviour of the Raman bands and the spectral background during tip retraction. The use of TERS as a single-molecule analytical tool is demonstrated in a concentration study on a triaryl dye in optical resonance with the incident laser line on Au(111). As a single-molecule analytical tool, TERS prevails over routinely employed fluorescence spectroscopy, because, at equally high photon count rates, no modification (labelling) of the sample molecule is needed and Raman fingerprint spectra provide much richer chemical information than fluorescence spectra. The high versatility of TERS is proven by probing target species of high biochemical relevance that are off resonance with the excitation laser line: The detection and identification of (sub)monolayers of the four DNA bases adsorbed at Au(111) is a first important step towards the investigation of DNA strands. To improve the TERS experiment, we have designed and constructed a new set-up with a parabolic mirror as light-focussing device. The new, much more compact design facilitates temperature and vibration insulation as well as control of the working environment. To employ TERS upon electrochemical conditions at controlled surface potentials in electrolyte and to adapt it for work in UHV were the main goals of carrying out large modifications of the existing set-up.||Das junge Forschungsgebiet der Plasmonik, das sich mit der Wechselwirkung von Licht mit oszillierenden Elektronen in Metalloberflächen oder -partikeln beschäftigt, gewinnt zunehmend an Bedeutung in der Oberflächenforschung. Das Konzentrieren von (sichtbarem) Licht auf nanometergrosse Bereiche durch Anregung lokalisierter Oberflächenplasmonen eröffnet völlig neue Forschungswege, da das Abbe-Limit von $lambda$/2, das die Auflösung des optischen Fernfeldes begrenzt, keine Rolle mehr spielt. Mit auf Plasmonenanregung basierender spitzenverstärkter Raman-Spektroskopie wurde ein langerwartetes Ziel in der Oberflächenforschung erreicht: Topographische und chemische Informationen über Adsorbat(sub)monolagen bis hin zu einzelnen Molekülen auf atomar flachen Substraten werden mit Hilfe der eleganten Kombination von Rastertunnelmikroskopie und Raman-Spektroskopie gleichzeitig gewonnen. In dieser Doktorarbeit werden verschiedene Ansätze bezüglich einer Verbesserung der physikalischen, chemischen und technischen Eigenschaften spitzenverstärkter Raman-Spektroskopie verfolgt. Experimente, in denen der Abstand zwischen Tunnelspitze und Substrat gezielt variiert wird, erlauben die Untersuchung der Nahfeldverteilung unterhalb der Tunnelspitze. Die beobachteten spektralen Änderungen von diskreten Ramanbanden und breitem Untergrund werden gezeigt und analysiert. Der Gebrauch spitzenverstärkter Raman-Spektroskopie zur Untersuchung von einzelnen Molekülen wird am Beispiel eines Triarylfarbstoffes, der in optischer Resonanz mit der Anregungswellenlänge liegt, demonstriert. Die Flexibilität der Methode wird mit der Analyse von kleineren Molekülen hoher biochemischer Relevanz, die nicht in Resonanz mit der Anregungswellenlänge liegen, aufgezeigt: Nachweis und Identifizierung von (Sub)Monolagen von DNA-Basen auf Au(111) sind ein erster wichtiger Schritt in Richtung Analyse ganzer DNA-Stränge. Eine veränderte Versuchsanordnung, in der ein Parabolspiegel anstelle einer Objektivlinse das einfallende Licht auf die Spitze fokussiert, wird beschrieben.
