Durch seine Beteiligung an der Biogenese der Typ 1-Fimbrien beeinflusst SurA zudem die Virulenz des uropathogenen E. coli-Zystitis-Isolates UTI89 (UPEC). In dieser Arbeit wurde am Beispiel der Pyelonephritis-Isolate 536 und CFT073 erstmals gezeigt, dass sich die pleiotropen Effekte von SurA auch auf weitere UPEC-Virulenzeigenschaften auswirken. In UPEC 536 und CFT073 führte das Fehlen von SurA, wie bereits für apathogene E. coli beschriebene Phänotypen bekannt, zu einem Defekt der äußeren Membran sowie zu einer konstitutiven Induktion der σE-abhängigen Stressantwort. Da das Proteom der Zelloberfläche in den von UPEC häufig in Assoziation mit Toxinen und Virulenzfaktoren sekretierten Außenmembranvesikeln (OMV) repräsentiert ist, wurde der Einfluss von SurA auf ihre Biogenese und Proteinzusammensetzung im Isolat 536 näher untersucht. Elektronenmikroskopische Untersuchungen zeigten, dass die 536-surA-Mutante vermehrt kleinere Vesikel im Vergleich zum Wildtyp abschnürt. Mutmaßlich wird dies durch die gestörte Membranintegrität und die Akkumulation von LPS im Periplasma hervorgerufen. Die Proteomanalyse der 536-OMV ergab, dass etwa 1/3 der detektierten Proteine durch SurA beeinflusst sind. Unter den Proteinen, die in den OMV der surA-Mutante in geringerer bzw. erhöhter Konzentration vorlagen, wurden die Flagellin-Untereinheit FliC und das PspA-Protein identifiziert. Damit wurde erstmals ein Zusammenhang zwischen SurA und der Motilität bzw. SurA und dem Psp-System, das in vielen Bakterien durch extrazytoplasmatischen Stress induziert wird, aufgezeigt. Die flagellenvermittelte Motilität ist für die Ausbreitung von UPEC im Wirt von zentraler Bedeutung. In dieser Arbeit konnte für die UPEC-Stämme 536, CFT073, UTI89 und F11 ein Motilitätsdefekt der surA-Mutanten auf Schwärmagarplatten nachgewiesen werden. EM-Aufnahmen der 536-surA-Mutante zeigten, dass 2/3 weniger Zellen intakte Flagellen trugen als der Wildtyp. Die Transkriptmengen von fliC und weiteren Genen der Flagellen-Regulationskaskade waren hingegen unverändert. Möglicherweise beruht der Motilitätsdefekt indirekt auf der verminderten Integrität der äußeren Membran und einer dadurch bedingt unzureichenden Verankerung der für den Flagellenaufbau notwendigen Hakenstruktur. Verringerte FliC-Level ließen sich durch eine negative Rückkopplung des resultierenden Assemblierungsdefekts auf die posttranskriptionale Regulation von FliC erklären, die bisher allerdings nicht beschrieben ist. Ferner konnten in dieser Arbeit erste Hinweise auf einen hämolytischen Defekt der 536-surA-Mutante mittels quantitativer Hämolysetests bestätigt werden. Filtrierte Überstände von Kulturen der surA-Mutante in der späten exponentiellen Wachstumsphase wiesen reproduzierbar eine signifikant geringere hämolytische Aktivität auf als die des Wildtyps. Die Expression von hlyA ist in der surA-Mutante unbeeinflusst. Immunochemische Analysen weisen aber auf eine erhöhte Sekretion von HlyA und eine rasche Degradation der sekretierten Toxinmoleküle in surA-defizienten Zellen hin. Schließlich wurde im Zuge dieser Studien erstmals nachgewiesen, dass das Fehlen von SurA zu einer erhöhten Aktivität von noch näher zu identifizierenden außenmembranassoziierten Phospholipasen führt.","The Escherichia coli periplasmic chaperone SurA for ß-barrel outer membrane proteins plays an integral role in cell envelope homeostasis. Due to its participation in the biogenesis of type 1 fimbriae SurA also influences the virulence of the uropathogenic E. coli cystitis isolate UTI89 (UPEC). In this study using the example of the pyelonephritis isolates 536 and CFT073 we showed for the first time that the pleiotropic effects of SurA affect further UPEC virulence factors. In UPEC 536 and CFT073 lack of SurA led to defects of the outer membrane and the constitutive induction of the σE-dependent stress response. Since the proteo-me of the cell surface is represented by the secreted outer membrane vesicles (OMV), which are often associated with toxins and virulence factors, we investi-gated the impact of SurA on their biogenesis and protein composition of the isolate 536. Electron microscopic analyses revealed that the 536 surA mutant produces an increased number of smaller vesicles compared to the wildtype. Presumably, this is caused by a defective outer membrane integrity and accumulation of lipopolysaccharides (LPS) in the periplasm. Proteome analysis of the 536 OMV showed that about one third of the discovered proteins are influenced by SurA. Among the proteins which appeared in lower or higher concentrations we identified the flagellin subunit FliC and the PspA protein. Hence, we revealed a relationship between SurA and motility respectively SurA and the Psp-system (induced by extracytoplasmic stress) first. Flagellum-mediated motility is important for UPEC to disseminate within the host. In this study, we verified a motility defect of the UPEC 536, UTI89, CFT073 and F11 surA mutants on soft agar plates. Electron microscopic analyses of the 536 surA mutant revealed that bacteria cells produce two third less intact flagella com-pared to the wildtype. Transcript level analyses of fliC and further genes of the flagellar regulation cascade remained unchanged. Potentially, the motility defect is indirectly based on the diminished integrity of the outer membrane and thus, essentially needed for proper assembly, a sufficient anchorage of the flagellar hook. A negative feedback mechanism on the post-transcriptional regulation of FliC by reason of the assembly defect could be an assertion of decreased FliC level. Furthermore, using quantitive hemolysis assays, we could confirm first evidences that lack of SurA in UPEC 536 lead to a hemolytic defect. Filtrated culture super- natants at late exponential phase of the surA mutant showed a significant lower hemolytic activity than the wildtype. The expression of hlyA is unaffected in surA mutant cells. However, immunoblot analysis of surA deficient cells suggests an increased secretion of HlyA and a rapid degradation of the secreted toxin mole-cules.
