Zu diesem Zweck wurde zunächst die Leistungsfähigkeit des in Deutschland noch wenig gebräuchlichen selektiven Festnährmediums CHROMagar™Vibrio beim Nachweis des Zielbakteriums überprüft. Weiterhin wurde das zur Anreicherung von Vibrio spp. übliche alkalische Peptonwasser (APW) modifiziert und damit speziell an die Anzucht von V. parahaemolyticus angepasst. Daneben wurden zwei quantitative Nachweisverfahren von Bakterien verglichen. Die erarbeitete APW-Modifikation und die sensiblere Quantifizierungsmethode wurden dann bei der Untersuchung kontaminierter Miesmuscheln angewendet. Im Ergebnis konnte festgestellt werden; dass CHROMagar™Vibrio als selektives Festnährmedium dem herkömmlichen TCBS-Agar beim Nachweis von V. parahaemolyticus überlegen war. Mit ihrer charakteristischen lila Farbe und irisierenden Rändern liessen sich V. parahaemolyticus-Kolonien auf CHROMagar™Vibrio von anderen relevanten Bakterien deutlich besser abgrenzen; was die Verwechslungsgefahr erheblich reduzierte. Die Sensitivität verbessert sich deutlich; wenn der irisierende Rand als weiteres charakteristisches Merkmal herangzogen wird. Bezüglich der verschiedenen Modifikationen des alkalischen Peptonwassers können die Ergebnisse folgendermaßen zusammengefasst werden: 1\. Die Konzentration des zugesetzten Tris-Puffers (0;1; 0;05 und 0;01M) nahm keinen Einfluss auf das Keimwachstum. 2\. Ein Zusammenhang zwischen Keimdichte und pH-Wert (pH 8;5; 8;0 und 7;5) bestand nicht. 3\. In T-APW (0;01M; pH 8;5) mit einem Kochsalzgehalt von 5% waren signifikant höhere Keimdichten nachzuweisen als in herkömmlichem APW bzw.T-APW (0;01M; pH 8;5). Zudem wurde das Wachstum der Nicht-Vibrio-Begleitflora in diesem Medium deutlich gehemmt. 4\. Eine Anhebung des Peptongehalts des Anreicherungsmediums führte zu keiner Steigerung der V. parahaemolyticus-Wachstumsrate in T-APW (0;01M; pH 8;5; 5%). 5\. Von allen Variationen überzeugte T-APW (0;01M; pH 8;5; 5% NaCl; 2% Arabinose) als Anreicherungsmedium am meisten. Die Überlegenheit des derart modifizierten alkalischen Peptonwasser zeigte sich insbesondere bei gleichzeitiger Anwesenheit von V. alginolyticus; einem der Hauptkonkurrenten des Zielbakteriums. Während dieser V. parahaemolyticus in herkömmlichem APW regelmäßig überwucherte; gelang der Nachweis des Zielkeimes in T-APW (0;01M; pH 8;5; 5% NaCl; 2% Arabinose) deutlich zuverlässiger. Gleichzeitig wurde mit diesem Medium die sonstige typische; aber hier unerwünschte marine Begleitflora stark verdrängt. Zur Quantifizierung geringer Mengen von V. parahaemolyticus in Miesmuscheln erwies sich das MPN-Verfahren als vorteilhaft. Dabei bot sich eine Kombination von T-APW (0;01M; pH 8;5; 5% NaCl; 2% Arabinose) und CHROMagar™Vibrio an. Weil V. parahaemolyticus auf diesem Festnährmedium im Gegensatz zum TCBS-Agar recht zuverlässig differenzierbar ist; konnte schon vor der biochemischen Identifikation eine Verdachtsdiagnose gestellt werden. Ebenso wurde durch die deutlich geringere Verwechslungsgefahr mit anderen Vibrio spp. der Umfang der zeit- und materialaufwändigen biochemischen Analyse erheblich reduziert. Mit dieser optimierten Methode zum quantitativen und qualitativen Nachweis von V. parahaemolyticus in zweischaligen Weichtieren sollen die vorgestellten Untersuchungen einen Beitrag zur Lebensmittelsicherheit leisten.","The work at hand intended to improve the present conventional methods of the qualitative and quantitative detection of V. parahaemolyticus in bivalve molluscs. For these purposes the efficiency of the selective solid medium CHROMagar™Vibrio for the detection of the target bacterium was tested; which has rarely been used in Germany so far. Furthermore the broth generally used for the enrichment of Vibrio spp. was modified in several ways in order to adapt it selectively for the accumulation of V. parahaemolyticus. In addition; two enumeration procedures for bacteria were compared. The product of the APW- modification and the more sensitive enumeration method was applied to the examination of contaminated mussels. As a result it could be observed that CHROMagar™Vibrio outclasses TCBS-agar as a selective solid medium detecting V. parahaemolyticus. With their characteristic violet colour and iridescent edges colonies of V. parahaemolyticus on CHROMagar™Vibrio were much easier distinguishable from other relevant bacteria and therefore reduced the likelihood of confusion. If the iridescent edge is also consulted as a further characteristic criterion the sensitivity is improved considerably. As far as the different modifications of the alkaline peptonewater are concerned the results can be summarised as follows: 1\. The concentration of the Tris-buffer which was added (0;1; 0;05 and 0;01) had no influence on the bacteria count. 2\. A correlation between germ-number and pH-value (pH 8;5; 8;0 und 7;5) could not be detected. 3\. Compared to the common APW as well as T-APW (0;01M; pH 8;5) significant higher germ-number could be observed in T-APW ( 0;01M; pH 8;5) with a NaCl-concentration of 5%. Furhtermore the growth of accompanying bacterial flora was significantly inhibited. 4\. Raising the peptone- concentration of the enrichment-broth however did not improve the bacterial count of V. parahaemolyticus in T-APW (0;01M; pH 8;5; 5% NaCl). Finally T-APW (0;01M; pH 8;5; 5% NaCl; 2% Arabinose) produced the best results. 5\. Out of those variations T-APW (0;01M; pH 8;5; 5% NaCl; 2% Arabinose) was the most satisfying one. The superiority of APW modified that way became particularly apparent when V. alginolyticus; the most important competitor of the target bacterium; was also present. Whereas V. alginolyticus tended to overgrow V. parahaemolyticus in conventional APW the detection of the target bacterium in T-APW (0;01M; pH 8;5; 5% NaCl; 2% Arabinose) was much more consistent. At the same time the accompanying bacterial flora was reliably suppressed at the same time. To quantify small amounts of V. parahaemolyticus in mussels the MPN- procedure proved to be advantageous. When performing this enumeration method the combination of T-APW (0;01M; pH 8;5; 5% NaCl; 2% Arabinose) and CHROMagar™Vibrio was favorable. Since V. parahaemolyticus could be differentiated rather reliably; a tentative diagnosis could be given before the biochemical identification procedure. Due to the reduced likelihood of confusion the extent of the time- and material-consuming biochemical analysis was also minimized considerably.
