Die Nanomedizin besitzt dabei großes Potential bei der Entwicklung neuer Therapieformen und bei der Diagnostik verschiedenster Krankheitsbilder. Eisenoxidnanopartikel beispielsweise werden heute bereits als Kontrastmittel in der Magnetresonanztomographie oder zur Thermotherapie von Gehirntumoren am Patienten verwendet. Trotz ihrer klinischen Anwendung und ihres Potentials für weiterführende Thera-piekonzepte ist nur wenig darüber bekannt, welche Faktoren auf welche Weise die intrazellu-läre Aufnahme von Eisenoxidnanopartikeln in Zellen beeinflussen und welche Internalisierungsrouten dazu genutzt werden. Erkenntnisse über die grundlegenden Phäno-mene der Interaktion von Nanopartikeln mit humanen Zellen könnten eine gezielte Entwicklung von klinisch anwendbaren Nanopartikeln für eine Vielzahl von Indikationen ermöglichen. In der vorliegenden Arbeit sollte untersucht werden, wie Eisenoxidnanopartikel mit Silica-schale (SiO2-Fe2O3) von Tumorzellen internalisiert werden. Dazu sollte zuerst qualitativ und quantitativ die intrazelluläre Aufnahme von SiO2-Fe2O3 Partikeln in Abhängigkeit ihrer Ober-flächenbeschaffenheit in murine (C3H RS1) und humane (BT20, HeLa) Tumorzellen in vitro betrachtet werden. Es sollte sowohl der Nachweis der intrazellulären Aufnahme erbracht als auch die Auswirkungen unterschiedlicher Oberflächenmodifikationen auf die Aufnahmekinetik sowie die Aufnahmemenge der Nanopartikel in die verwendeten Zellen untersucht werden. In einem zweiten Schritt sollte gezeigt werden, welche Aufnahmemechanismen an der Inter-nalisierung ausgewählter SiO2-Fe2O3 Nanopartikel beteiligt sind. Durch den Vergleich von mindestens zwei unterschiedlichen Zelllinien sollte die Ableitbarkeit allgemeiner Tendenzen für Tumorzellen geprüft werden. Im dritten Teil der Arbeit sollte die potentielle Toxizität der verwendeten Nanopartikel beleuchtet werden. Die Arbeitshypothese lautete, dass die ver-stärkte intrazelluläre Anreicherung von SiO2-Fe2O3 Partikeln die Funktion der Endo- bzw. Lysosomen beeinträchtigen und dadurch zur Induktion von Autophagie führen könnte. Zur Untersuchung des Einflusses von Oberflächenmodifikationen wurden SiO2-Fe2O3 Partikel aus einer Charge verwendet, die entweder mit Thiol-, Carboxy-, Epoxy-, Amino-, PEG- oder Amino/PEG- Gruppen funktionalisiert worden waren. Die intrazelluläre Aufnahme in C3H RS1, BT20 und HeLa Zellen wurde mittels Transmissionselektronenmikroskopie (TEM) qualitativ untersucht. Die quantitativen Daten wurden durch die analytische Bestimmung des int- razellulären Eisengehaltes mittels Photometrie oder Massenspektrometrie erhoben. Für die Experimente zum Aufnahmemechanismus wurden die Zellen mit spezifischen siRNAs trans-fiziert, um die Expression bekannter Proteine aus der Endocytosemaschinerie zu verhindern (Caveolin-1, Clathrin, Flotillin-1, Dynamin2, PIP5Kα, GRAF1, CtBP1, CDC42). Die Auswirkungen auf die Nanopartikelaufnahme wurden entweder durch die erwähnte Eisenanalytik oder in Kombination mit fluoreszenzmarkierten Partikeln mittels konfokaler Fluoreszenzmikroskopie quantifiziert. Zur Untersuchung der Toxizität wurde die Expression des Markerproteins LC3B für Autophagosomen in HeLa Zellen bestimmt, welche unterschiedlichen Konzentrationen von SiO2-Fe2O3 Partikeln ausgesetzt waren. Die TEM-Aufnahmen zeigten, dass alle getesteten Partikel in das endosomale System von C3H RS1, BT20 und HeLa Zellen aufgenommen wurden. Es konnten keine Nanopartikel beobachtet werden, die sich frei im Cytosol oder im Zellkern befunden hätten. Die quantitative Eisenanalytik zeigte, dass positiv geladene aminofunktionalisierte Partikel mit kombinierter PEGylierung bei C3H RS1 Zellen (9,16±1,97 pg Fe/Zelle) und positiv geladene aminofunktionalisierte Partikel bei BT20 Zellen (55±4,7 pg Fe/Zelle) am stärksten zellassoziiert vorlagen. Am geringsten war der intrazelluläre Eisengehalt jeweils bei den negativ geladenen unmodifizierten und thiolisierten Partikeln. Mit Ausnahme der aminofunktionalisierten Partikel war die maximale Aufnahmemenge der getesteten Modifikationen innerhalb 48 h Inkubationszeit in beiden Zelllinien vergleichbar. Der größte Einfluss auf die Partikelaufnahme nach Transfektion mit spezifischen siRNAs konnte beobachtet werden, wenn die Proteine Caveolin-1 und CDC42 in HeLa Zellen depletiert waren. Im Falle von Caveolin-1 lag die intrazelluläre Eisenmenge je nach verwendeter Partikelart 23-41 % unter dem Wert der Kon-trollzellen. Zellen ohne CDC42 nahmen 46 % weniger PEGylierte SiO2-Fe2O3 Partikel intra-zellulär auf. Die Internalisierung von PEGylierten SiO2-Fe2O3 Partikeln wird bei HeLa Zellen also zu einem großen Teil durch Caveolin-1 und CDC42 vermittelt. Die Ergebnisse konnten jedoch nicht auf BT20 Zellen übertragen werden. Die Quantifizierung von LC3B bei HeLa Zellen nach Inkubation mit unterschiedlichen Konzentrationen von PEGylierten SiO2-Fe2O3 Partikeln zeigte eine konzentrationsabhängige Steigerung und somit eine vermehrte Bildung von Autophagosomen, die durch die Nanopartikelexposition verursacht wurde. Sowohl die intrazelluläre Lokalisierung der verwendeten Partikel als auch die ladungsabhän-gige Internalisierung bestätigen vergleichbare Befunde mit andersartigen Nanopartikeln aus der Literatur. Die Ergebnisse leisten dabei einen Beitrag zur Bewertung der Anwendbarkeit der getesteten Partikelspezies in vivo. Die Caveolin- und CDC42- abhängige Aufnahme von PEGylierten SiO2-Fe2O3 Partikeln durch HeLa Zellen wird hier das erste Mal beschrieben. Die beobachtete Autophagieinduktion zeigt, dass Eisenoxidnanopartikel, anders als zumeist in der Literatur berichtet wird, Einfluss auf den Zellstoffwechsel nehmen können. Dieser Effekt könnte auch therapeutischen Nutzen haben. Um die gewonnenen Erkenntnisse zu vertiefen, müssten zukünftig Studien mit weiteren Zelllinien und Primärzellen unterschiedlicher Gewebsidentität sowie mit verbesserten Nanopartikelsuspensionen durchgeführt werden.","Nanotechnology deals with materials and components, which are approximately between 1 and 100 nanometers in size. Nanomedicine has great potential for the developing of new forms of therapy and in the diagnosis of various diseases. Iron oxide nanoparticles, for ex-ample, are already being used as contrast agents in magnetic resonance imaging or thermal therapy of brain tumors in patients. Despite their clinical use and their potential for further therapy concepts, little is known about the factors that influence intracellular uptake of iron oxide nanoparticles into cells and which routes of cell entry are used. Insight of the basic phenomena of the interaction of nanoparticles with human cells could allow targeted devel-opment of clinically applicable nanoparticles for a variety of indications. In the present work should be explored, how iron oxide nanoparticles with silica shell (SiO2-Fe2O3) are internalized by tumor cells. First of all, the intracellular uptake of SiO2-Fe2O3 parti-cles should be measured qualitatively and quantitatively in murine (C3H RS1) and human (BT20, HeLa) tumor cells in vitro depending on their surface properties. The intracellular up-take of nanoparticles should be proven as well as the effects of different surface modifica-tions on the uptake kinetics and the amount of nanoparticles inside the cells used. In a second step should be shown, which uptake mechanisms are involved in the internalization of selected SiO2-Fe2O3 nanoparticles. By comparing at least two different cell lines, possible general trends should be deduced for tumor cells. In the third part, the potential toxicity of nanoparticles used should be investigated. It was hypothesized, that increased intracellular accumulation of SiO2-Fe2O3 particles could impair the function of endo- or lysosomes, thereby leading to the induction of autophagy. SiO2-Fe2O3 particles were used from the same batch to study the influence of surface modifi- cations. Therefore, the particle surface had been functionalized with thiols, carboxy groups, epoxy groups, amino groups, PEGs, or a combination of amino/PEG. The intracellular uptake in C3H RS1, BT20, and HeLa cells was investigated qualitatively by transmission electron microscopy (TEM). The quantitative data were collected through the analytical determination of the intracellular iron content by photometry or spectrometry. To investigate the uptake mechanisms, the expression of known proteins from the endocytotic machinery was knocked down by use of specific siRNAs in transfected target cells. These proteins were caveolin-1, clathrin, Flotillin-1, Dynamin2, PIP5Kα, GRAF1, CtBP1, and CDC42. The effects on particle uptake were quantified either by determination of the intracellular iron content or by confocal fluorescence microscopy in combination with fluorescence-labeled particles. To investigate the toxicity of SiO2-Fe2O3 particles, the expression of the marker protein LC3B for autophagy was measured in HeLa cells, which were exposed to different concentrations of nanoparti-cles. The TEM images showed that C3H RS1, BT20, and HeLa cells internalized all tested particles. They were located in vesicles of the endosomal system in the cytoplasm of the cells. No free nanoparticles could be observed in the cytosol or in the nucleus. Quantitative iron analysis demonstrated that positively charged amino-functionalized particles with combined PEGylation showed the best affinity to C3H RS1 cells (9.16±1.97 pg Fe/cell). The positively charged amino-functionalized particles were associated with BT20 cells (55 ±4.7 pg Fe/cell) to the highest level compared to all the other tested modifications. The lowest intracellular iron content was observed for negatively charged unmodified and thiolated particles in both cell lines. With the exception of the amino-functionalized particles, the maximum uptake level of the tested modifications in both cell lines was comparable within 48 h incubation. The biggest influence on particle uptake after transfection with specific siRNAs could be observed during the depletion of caveolin-1 and CDC42 in HeLa cells. Depending on the particle type used, knockdown of caveolin-1 decreases intracellular iron content about 23-41 % compared to control cells. Cells lacking CDC42 internalize 46% less PEGylated SiO2-Fe2O3 particles. Hence the uptake of PEGylated SiO2-Fe2O3 particles in HeLa cells is primarily mediated by caveolin-1 and CDC42. However, the results could not be transferred to BT20 cells. The quantification of LC3B in HeLa cells after incubation with different concentrations of PEGylated SiO2-Fe2O3 particles showed a concentration-dependent increase and thus an increased formation of autophagosomes, which was caused by the exposure to nanoparticles. Both the intracellular localization of the particles and the charge-dependent internalization confirm comparable results with different nanoparticles in the literature. The results of this study contribute to the evaluation of the tested particle species for applications in vivo. The caveolin-1- and CDC42-dependent uptake of PEGylated SiO2-Fe2O3 particles by HeLa cells is described here for the first time. The observed induction of autophagy shows that iron oxide nanoparticles with silica shells could influence the cell metabolism, which is contrary to most findings reported in literature. This effect could be useful in therapy.
