Da der Trend zu immer kleineren aber gleichzeitig leistungsfähigeren Geräten und Instrumenten geht, deren Bestandteile mit den gegenwärtigen Methoden nur schwer herzustellen sind, erhoffen sich speziell die Materialwissenschaften durch Kombination von organischen und anorganischen Molekülen die gezielte Synthese von submikrometergroßen Hybridmaterialien mit neuartigen optischen, elektronischen und katalytischen Eigenschaften. Ein entscheidender Faktor ist dabei die Wahl eines passenden Templats, welches die für die Interaktion notwendigen Liganden im Raum präsentiert. Ein solches Templat stellt das coiled coil Faltungsmotiv dar, das eine gerichtete Präsentation einer Vielzahl von Liganden bei vollständiger Kontrolle über deren Abstand untereinander ermöglicht. Gleichzeitig erlaubt die chemische Variabilität der Aminosäureseitenkette eine Anwendbarkeit für eine Vielzahl von Fragestellungen. Im ersten Teil dieser Arbeit werden die Auswirkungen einer räumlich unterschiedlichen Präsentation geladener Aminosäuren in einem coiled coil Peptid auf die Organisation von Gold-Nanopartikeln mittels attraktiver elektrostatischer Anziehungskräfte untersucht. Es konnte gezeigt werden, dass ein bestimmtes Verhältnis aus präsentierten geladenen Aminosäuren zur Peptidlänge ein entscheidendes Kriterium für die Organisation der Nanopartikel darstellt. Darüber hinaus wurde aufgezeigt, dass die Quartärstruktur des Peptids sowohl eine entscheidende Rolle bei der Struktur der gebildeten Peptid-Nanopartikel-Aggregate spielt als auch einen großen Einfluss auf die Thermodynamik der Interaktion besitzt. Im zweiten Teil dieser Arbeit wurde das coiled coil Faltungsmotiv für die Strukturaufklärung verschiedener Tandem SH2-Domänen genutzt. Das Peptiddesign wurde dahingehend modifiziert, dass eine selektive Bindung von zwei phosphorylierten Peptidliganden ermöglicht wurde. Die hier präsentierten Ergebnisse zeigten, dass sowohl eine definierte räumliche Ausrichtung der Liganden als auch ein definierter Abstand der zwei Liganden für eine ideale Bindung an die Tandem SH2-Domänen notwendig war.","The concept of multivalent interactions, which is borrowed from nature, offers the advantage of dramatically enhanced binding and thus plays a key role in solving a variety of problems in biochemistry and medicine, as well as in the fields of supramolecular chemistry and material science. In spite of the increasing demand for even smaller but more powerful devices and instruments, their components are difficult to manufacture with current methods. Therefore, especially materials science tries to synthesize hybrid materials in the submicrometer range by combining organic and inorganic building blocks with novel optical, electronic and catalytic properties. Thus, a crucial factor is the choice of an appropriate template, which presents the interacting ligands in a directed manner. The coiled-coil folding motif allows for the directed presentation of a variety of ligands with complete control over the distances between these ligands. At the same time, the chemical variability of amino acid side chains offers numerous control mechanisms. In the first part of this thesis, the effects of altering the formal charge of amino acids in a coiled- coil peptide for the organization of gold nanoparticles by means of attractive electrostatic interactions was investigated. It was shown that a certain ratio of charged amino acids per unit peptide length is crucial for the organization of nanoparticles. In addition, the quaternary structure of the peptide plays an important role in the morphology of the formed peptide-nanoparticle aggregates as well as on the thermodynamics of the reaction. In the second part of this thesis, the coiled coil folding motif was used for the structure elucidation of various tandem SH2 domains. The peptide design was modified for the selective attachment of two phosphorylated peptide ligands in the f-position of the coiled coil at different distances.
