Getestet wurde, ob die unzureichende Migration von tumorspezifische T-Zellen in den Tumor (1), die Anergie der Tumor infiltrierenden T-Zellen (2), ein terminales und nicht mehr funktionelles Differenzierungsstadium der T-Zellen im Tumor (3), die Präsenz von regulatorischen T-Zellen im Tumor (4), Coinhibtion der T-Zell-Antworten im Tumor (5) oder die direkte Blockade der T-Zell-Antwort durch Kontakt mit den Tumorzellen (6) verantwortlich sind. In den untersuchten Tumorproben konnte ein großer Teil der Hypothesen widerlegt werden. Lediglich für die Relevanz der Coinhibition und die direkte Wirkung der Tumorzellen auf T-Zellen wurden Belege gefunden. Ingesamt zeigte sich eine Infiltration und Anreicherung von tumorspezifischen T-Zellen im Tumor, die mit einer tumorspezifischen Aktivierung einher ging. Weiterhin ergaben sich Hinweise darauf, dass die teilweise mit dem Überleben der Patienten korrelierte Expression coinhibitorischer Rezeptoren nicht notwendiger Weise Ursache, sondern eher Ausdruck der Dysfunktionalität der T-Zellen ist.","The aim of this work was to test different hypotheses for the insufficient anti tumor immunity in human melanoma and lymphoma samples, by analyzing phenotype and properties of tumor-infiltrating T cells and T cell from peripheral blood of patients and healthy donors. It was tested wether insufficient migration into the tumor (1), anergy of tumor-infiltrating T cells (2), terminal and nonfunctional differentiation state of T cells infiltrating the tumor (3), presence of regulatory T cells (4), expression of coinhibitory receptors (5) or direct blockade of T cell responses mediated by tumor cell contact were the cause for the dysfunctionality of T cells in the tumor. In the Samples analyzed, most of the hypotheses were refuted. Only coinhibition and direct contact with the tumor cells seem to be of relevance for the local immune suppression in the tumormicroenvironment. All in all an infiltration and enrichment of tumor-specific T cells in the tumor could be shown and was accompanied with tumor-specific activation.
