Heidrun Fink","n","2007-03-13","2018-06-07T16:23:11Z","2007-05-24T00:00:00.649Z","2007-05-31","2007","Einleitung: In dieser Arbeit wurden in der `Whole Embryo Culture´ (WEC) N-Methyl-2-pyrrolidon (NMP) und seine Metaboliten auf Embryotoxizität untersucht. Die WEC eig-net sich unter bestimmten Umständen für bestimmte Substanzgruppen gut für die Ab-klärung von Wirkmechanismen und für die Testung auf eine spezifische Embryotoxizi-tät. Zusätzlich wurde das `Whole-immuno- staining´ (WIS) der Neuralleistenzellen (NCC) und der Gehirnnerven durchgeführt, um beobachtete Defekte auf zellulärer E-bene nachvollziehen zu können. Material und Methoden: Für die WEC wurden Rattenembryonen am Gestationstag 9,5 sowohl in Kulturmedium (als Lösungsmittelkontrolle), als auch in Testansätzen mit unterschiedlichen Konzentrationen von NMP und seinen drei Metaboliten inkubiert. Sie wurden unter einem definierten Gaspartialdruck und bei 38,5°C für 48 Stunden in ei-nem rotierenden System kultiviert. Für das WIS wurden Embryonen aus der WEC mit primärem Antikörpern inkubiert: Der CRABP-I-Antikörper (zelluläres Retinsäure bin-dendes Protein 1) markierte die NCCs und der 2H3-Neurofilament-Antikörper die Ge-hirnnerven. Durch den Peroxidase-gekoppelten sekundären Antikörper wurde der Farbstoff Naphthol in ein blaues Präzipitat umgewandelt. Ergebnisse: Für alle Substanzen wurden in der WEC konzentrationsabhängige Effek-te beobachtet. Bei NMP traten vor allem Abnormitäten am Kopf auf. Dieser war unpro-portional verkleinert, das Auge erschien dorsoventral abgeflacht, es bestand eine Ad-häsion des zweiten Kiemenbogens mit dem ersten und seiner Umgebung. Neben die-sen Abnormitäten wurde eine Veränderung in Form einer Umfangsvermehrung in Höhe des Trigeminalganglions beobachtet. Diese konnte mit Hilfe des morphologischen Sco-ring-Systems nicht bewertet werden und die Einschätzung als Abnormität war deshalb nicht möglich. Beim WIS der 2H3-Neurofilamente wurden Retardierungen und Anoma-lien der kranialen Nerven in dieser Kopfregion bei Embryonen exponiert mit NMP fest-gestellt. Bei dem WIS mit CRABP-I war es nicht möglich eine Störung der NCC-Wanderung durch NMP im Vergleich zur Kontrollsituation festzustellen. Diskussion: In der WEC wurde für NMP und den Metabolit 5-Hydroxy-N-Methyl-2-pyrrolidon (5H-NMP) ein schwach embryotoxisches Potential festgestellt, welches sich in spezifischen morphologischen Abnormitäten bei den Embryonen manifestierte. Die Metaboliten N-Methylsuccinimid (MSI) und 2-Hydroxy-N-Methylsuccinimid (2H-MSI) wurden als nicht embryotoxisch klassifiziert. Sie verursachten nur einen generellen embryotoxischen Effekt bei sehr hohen unphysiologischen Konzentrationen. Da die im WIS detektierten Veränderungen an den Gehirnnerven der Embryonen inkubiert in NMP weder in der Kontrollsituation noch bei Ex-vivo-Embryonen auftraten, gab das WIS einen Hinweis auf eine spezifische Neurotoxizität von NMP. Schlussfolgerung: NMP scheint die für die Embryotoxizität in vivo verantwortliche Substanz zu sein. Die Ergebnisse des WIS lieferten im Fall von NMP ergänzende In-formationen zu den morphologischen Veränderungen und konnten zu diesen in einen sinnvollen Zusammenhang gestellt werden. The WEC is suited to scrutinise the mode of action and the specific embryotoxicity. In addition the whole-immuno-staining (WIS) of the neural crest cells (NCCs) and the cranial nerves was performed to understand defects observed in the WEC on a cellular level. Materials and methods: For the WEC 9.5-days-old rat embryos were incubated as control embryos and as test groups of increasing concentrations of NMP and its me-tabolites. They were cultured under defined conditions in a rotating system for 48 hours. For the WIS embryos of the WEC were treated with primary antibodies: the CRABP-I-antibody (cellular retinoic acid binding protein 1) was used to detect the NCCs, the 2H3-neurofilament-antibody to detect the cranial nerves. Naphthol was transformed into a blue precipitate by the peroxidase of the secondary antibody. Results: The substances induced concentration-dependent effects in the WEC. NMP caused abnormalities particularly in the head region. In proportion to the body the head was smaller, the eye seemed to be flattened in its dorsoventral axis and the second branchial arch was adhered to the first and to its surroundings. Beside these abnor-malities there was an alteration which occurred as a prominent node in the area of the trigeminal ganglion. This observation could not be judged with the scoring system used and therefore it was impossible to classify it as a abnormality. The WIS of the 2H3-neurofilament showed retardations and anomalies in the development of cranial nerves in embryos exposed to NMP. Using the WIS with CRABP-I it was not possible to de-termine an altered mirgration of NCCs after treatment with NMP in comparison to the situation in the control group. Discussion: In the WEC, a weak embryotoxic potential of NMP and the metabolite 5-Hydroxy-N-Methyl-2-pyrrolidone (5H-NMP) was determined which was manifested in specific morphological malformations of the embryos. The metabolites N-Methylsuccinimid (MSI) and 2-Hydroxy-N-Methylsuccinimid (2H-MSI) were classified as non embryotoxic, because they caused only general embryotoxic effects at very high non physiological concentrations. The observed effects on the cranial nerves after ex-position to NMP were not seen in the control situation nor in ex-vivo-embryos. So the WIS provides an indication of a specific neurotoxicity of NMP in vitro. Conclusion: NMP seemed to be the responsible substance for the embryotoxicity ob-served in in-vivo-studies. The results of the WIS provided amendments on morphologi-cal changes seen in the WEC and were linked with them.
