Die Apoptose spielt eine wichtige Rolle in der angeborenen Immunabwehr und wird über vielfältige Stimuli induziert. Mitochondrien sind für die Apoptose von großer Bedeutung, da hier die Signale mehrerer Apoptosewege zusammenführen. Hierbei entscheidet das Verhältnis von antiapoptotischen (Bcl-2, Bcl-xL) zu proapoptotischen (z.B. Bak, Bax) Proteinen der Bcl-2 Familie über das Leben der Zelle. Um Mitochondrien- vermittelte Apoptose zu inhibieren exprimieren viele Viren Bcl-2-Homologe, die sowohl Bak- als auch Bax-mediierte Apoptose blockieren. Cytomegalieviren der Mäuse und Ratten exprimieren kein Bcl-2 Homolog, verfü-gen jedoch über einen Bax-spezifischen Inhibitor (vMIA). Zu Beginn dieser Arbeit war bekannt, dass die Deletion der m41 Genregion aus dem MCMV-Genom zu Apoptose-Induktion in infizierten Zellen führt. Ziel dieser Studie war es herauszufinden, auf welche Art das verantwortliche Protein die Apoptose inhibiert, und ob es dazu weitere virale Proteine benötigt. Die Ergebnisse der Arbeit zeigten, dass der m41 Lokus zwei unterschiedliche Proteine kodiert. Das größere m41 Protein besitzt ein Molekulargewicht von etwa 20 kDa und ist im Golgi lokalisiert. Innerhalb des m41 Gens liegt in einem anderen Leserahmen das Gen für das m41.1 Protein. Dieses ist etwa 10 kDa groß und weist eine mito-chondriale Lokalisierung auf. Durch Konstruktion von Virusmutanten, die nur eines der bei-den Gene exprimieren, konnte gezeigt werden, dass m41 bei den hier untersuchten Bedingun-gen nur eine untergeordnete Rolle spielt. Lediglich in infizierten Makrophagen war ein Phä-notyp zu beobachten Von größerer Relevanz erwies sich das mitochondrial lokalisierte m41.1 Protein. Es konnte nachgewiesen werden, dass m41.1 die Oligomerizierung des durch die Infektion aktivierten Bak verhindert und dadurch die Auslösung der Apoptose inhibiert. Die anti- apoptotische Funktion von m41.1 ist richtet sich spezifisch gegen Bak, ein Einfluss auf die Bax-vermittelte Apoptose war nicht nachzuweisen. Weiterhin konnte gezeigt werden, dass für die Funktion von m41.1 kein weiteres virales Protein benötigt wird, und dass dieser Apop-toseinhibitor in den Cytomegalieviren der Mäuse und Ratten konserviert ist. Cytomegaloviren inhibieren Apoptose an der mitochondrialen Schaltstelle auf unkon-ventionelle Art und Weise. Im Gegensatz zu den meisten Viren exprimieren sie kein Bcl-2-Homolog, sondern inhibieren Bax und Bak mit zwei unterschiedlichen Proteinen (vMIA und m41.1). Dieser bemerkenswerte Mechanismus der Apoptose- Inhibition wurde bislang bei keinem anderen Pathogen beschrieben.","Due to their intracellular replication, cytomegaloviruses need to inhibit the induction of apoptosis in infected cells until progeny viruses are produced. Apoptosis is an important part of the innate immune system and can be induced by multiple stimuli. Since various path-ways converge at mitochondria, these organelles are of major importance for apoptosis. The balance of antiapoptotic (Bcl-2, Bcl-xL) and proapoptotic (Bak, Bax) Bcl-2 family proteins decides between death and survival of the cell. Many viruses express Bcl-2 homologues to inhibit mitochondria-mediated apoptosis via Bax and Bak. However, murine and rat cy-tomegaloviruses do not express such proteins. Instead they code for a Bax-specific inhibitor of apoptosis, which has been named vMIA Previous studies identified an antiapoptotoc function of the m41 gene region because the deletion of this gene lead to apoptosis of infected cells. The aim of this work was to iden-tify the mechanism of apoptosis inhibition used by the responsible proteins and to find out whether additional viral proteins are needed for this function. This study shows that the m41 locus codes for two separate proteins. The larger m41 protein has a molecular weight of ap- proximately 20 kDa and localises to the Golgi. The smaller 10 kDa m41.1 protein is encoded within the m41 gene using a different reading frame and localises to mitochondria.By con-structing virus mutants expressing only one of the two proteins it could be shown that m41 is of minor importance in the settings used. The only phenotype observed by a virus missing the m41 protein was a reduced viability of infected macrophages. Instead the mitochondrial local-ised m41.1 is of major importance. It could be demonstrated that m41.1 inhibits the oligomer-isation of activated Bak thereby inhibiting the onset of apoptosis. Furthermore, the antiappop-totic function of m41.1 is Bak-dependent because an influence on Bax-mediated apoptosis is missing. Additionally, the function of m41.1 is independent of other viral proteins and ho-mologous apoptosis inhibitors are conserved in murine and rat cytomegaloviruses. Cytomegaloviruses inhibit apoptosis at the mitochondrial checkpoint in an unconven-tional manner.
