In Hefe spielt dabei der HRD-Komplex eine zentrale Rolle. Dieser Ubiquitinligase- Komplex besteht aus der RING-Finger E3-Ubiquitinligase Hrd1 und ihrem Interaktionpartner Hrd3, Der1, das durch Usa1 rekrutiert wird, und Yos9. Das Substrat-erkennende Protein Yos9 bindet auf der luminalen Seite des Komplexes an die luminale Domäne des Membranproteins Hrd3. Hrd3 interagiert mit ERAD- Substraten unabhängig von ihrem Glykosylierungsstatus, während Yos9 spezifische Oligosaccharidstrukturen gebundener Substrate erkennt, die eine Fehlfaltung signalisieren. Ziel dieser Arbeit war die Untersuchung der Protein-Protein-Interaktionen zwischen Usa1 und Hrd1 sowie zwischen Yos9 und Hrd3. Darüber hinaus sollten Einblicke in die molekulare Struktur dieser Proteine gewonnen werden. Die direkte Interaktion zwischen den cytosolischen Domänen von Usa1 und Hrd1 wurde mit biophysikalischen Methoden in vitro nachgewiesen. Die Proteine assoziieren in einer 1:1-Stöchiometrie mit einer Dissoziationskonstante im oberen nanomolaren Bereich. Der Oligomerisierungszustand des Komplexes konnte nicht zweifelsfrei bestimmt werden. Während die Masse des Komplexes im statischen Lichtstreu-Experiment auf eine 2+2-Assoziation hindeutet, entspricht sie in der analytischen Ultrazentrifuagtion einer 1+1-Assoziation. Vor kurzem wurde eine Selbst- Assoziation von Usa1 und Hrd1, basierend auf ihren cytosolischen Domänen, nachgewiesen. Vor diesem Hintergrund scheint der 2+2-Assoziationsmodus der Realität in der Zelle näherzukommen, dies sollte jedoch durch weitere Untersuchungen validiert werden. Die Kristallstruktur der postulierten Hrd3-Bindedomäne von Yos9, nachfolgend DD (Dimerisierungs-Domäne) benannt, wurde bei einer Auflösung von 2,5 Å bestimmt. Basierend auf der Dimerisierung der DD in der Struktur wurde die Selbst-Assoziation von Yos9 untersucht. Die Dimerisierung der Yos9-DD und des Volle-Länge-Proteins in Lösung wurde durch analytische Ultrazentrifugation nachgewiesen. Mutagenese-Studien bestätigten eine übereinstimmende Dimer-Anordnung im Kristall und in Lösung. Zusätzliche biochemische Untersuchungen konnten eine Beteiligung der Yos9-DD an der Interaktion zu Hrd3 nicht bestätigen. Stattdessen wurde die Interaktion in vitro durch den N-terminalen Bereich von Yos9 inklusive der MRH-Domäne (aa 24-262) vermittelt. Die Struktur der Yos9-DD stellt den ersten strukturellen Einblick in die funktionell wenig charakterisierten Domänen von Yos9 dar. Darüber hinaus wurde hier erstmalig eine Selbst-Assoziation auf der luminalen Seite des HRD-Komplexes beschrieben. Die Dimerisierung von Yos9 unterstützt damit das Modell eines dimeren HRD-Komplexes.","In the endoplasmic reticulum (ER), misfolded proteins are targeted for degradation by a pathway frequently called ER-associated protein degradation (ERAD). In yeast, the HRD complex plays a central role in this pathway. This ubiquitin ligase complex consists of the RING-finger E3 ubiquitin ligase Hrd1 and its binding partner Hrd3, Der1, which is recruited by Usa1, and Yos9. The recognition factor Yos9 resides on the luminal side of the complex, where it binds to the luminal domain of the membrane anchored Hrd3. Hrd3 interacts with ERAD substrates regardless of their glycosylation state, while Yos9 scans bound substrates for glycan structures that indicate misfolding. This thesis aimed at elucidating protein-protein interactions between Usa1 and Hrd1 and between Yos9 and Hrd3, as well as gaining insights into the molecular structure of these components. A direct interaction between the cytosolic domains of Usa1 and Hrd1 was demonstrated in vitro using biophysical methods. The proteins interact in a 1:1 stoichiometry with a dissociation constant in the upper nanomolar range. Data concerning the oligomeric state of the complex are inconsistent, with static light scattering indicating a 2+2 association mode, while analytical ultracentrifugation indicates a 1+1 association. As both Usa1 and Hrd1 were recently shown to self-associate based on their cytosolic domains, the 2+2 association mode is more likely to represent the situation within the cell, but this still has to be validated. The crystal structure of the postulated Hrd3 binding domain of Yos9, here denoted DD (dimerization domain), was analyzed at 2.5 Å resolution. Based on the dimeric arrangement in this structure, the self-association of Yos9 was studied. Analytical ultracentrifugation demonstrated the dimeric assembly of both the DD and full-length Yos9 in solution. Mutagenesis studies confirmed that the dimeric arrangement in the crystal structure is consistent with the arrangement in solution. Additional biochemical assays indicated that the Yos9-DD does not confer binding to the HRD complex, as suggested by previous experiments. Instead, the N-terminal part of Yos9 including the MRH domain (aa 24-262) mediated the interaction in vitro. The Yos9-DD structure represents the first structural insight into the sparsely annotated regions of Yos9. Furthermore, this is the first time that self-association on the luminal side of the HRD complex has been reported.
