Bezüglich Rezidivfreiheit und Überlebenswahrscheinlichkeit präsentieren sich aktuell bei der Lebertransplantation die besten Ergebnisse. Posttransplantär ist eine lebenslange Immunsuppression mit Calcineurininhibitoren (CNI) und/oder Antimetaboliten wie Sirolimus und Mycophenolatmofetil (MMF) notwendig. Für letztere ist bei verschiedenen Malignomentitäten ein antiproliferativer Effekt bekannt. Methodik: Ziel dieser Arbeit war die Testung der mono- und dualtherapeutischen Proliferationshemmung von Sirolimus und MMF auf die HCC- Zelllinien SKHEP1 und HEP3B. Nachgewiesen wurde die Reduktion der Zellviabilität in vitro durch den XTT -Proliferationstest. Ergebnisse: Die ausgeprägteste Hemmung erreichte Sirolimus bei SKHEP1 mit einer Zellviabilität von 9,7 % (p = 0,004). Die Zelllinie HEP3B wurde durch die duale Anwendung maximal gehemmt auf 26,4 % (p = 0,004). Eine Dosisreduktion war jeweils durch die duale Anwendung möglich. Schlussfolgerung: Es zeigte sich eine inhibierende Wirkung von Sirolimus und MMF auf die HCC-Zellen SKHEP1 und HEP3B. Somit ist klinisch eine antitumorale Wirkung denkbar. Indikationen wären der Einsatz zur Rezidivprophylaxe, zur Reduktion des Zweitkarzinomrisikos, zur Verhinderung von de novo-Malignomen sowie die palliative Therapie.","Hepatocellular carcinoma (HCC) is one of the most common malignancies worldwide and is associated with a low cure rate. Currently liver transplantation represents the best results concerning the recurrence rate and the post operative survival. A lifelong immunosuppression is required with calcineurin inhibitors (CRI) and/or antimetabolites such as sirolimus and mycophenolate mofetil (MMF). For sirolimus and MMF an antiproliferative effect is proven with various tumor celllines. Methodology: The aim of this study was to detect the mono and dual therapeutic inhibition of proliferation by sirolimus and MMF on the HCC cell lines HEP3B and SKHEP1. The effect of these drugs on the cell lines was tested in vitro by the XTT proliferation assay. Results: The strongest inhibition of proliferation was achieved by sirolimus on SKHEP1 with a cell viability of 9,7 % (p = 0,004). The viability of the HEP3B-cell line was reduced to 26,4 % (p = 0,004) with a dual application of the drugs. A dose reduction was possible with dual application. Conclusion: The results presented an inhibitory effect of sirolimus and/or MMF on the HCC- cell lines SKHEP1 and HEP3B. Due to the combined use of sirolimus and MMF an additional anti-tumor effect is conceivable in clinical settings.
