Folgende Ergebnisse ergaben sich: 1\. Die Sequenzierung des Matrixproteins bei verschiedenen Stämmen zeigt eine starke phylogenetische Homologie. Die Resultate bestätigten eine hohe erneute Stabilität des M Proteins bei der Vermehrung von Bornaviren in Zellen und eine starke Konservierung dieses Matrix Proteins in den Virusisolaten von Mensch und Tier. 2\. Das Gen des Matrixproteins wurde in einen prokaryotischen Expressionsvektor kloniert, das das Matrixprotein in Bakterien (E. coli) überexprimiert und durch Immobilized Metal Affinity Chromatography (IMAC) stark gereinigt wurde. Dieses Protein fand nun Anwendung als Antigen zur Detektion von spezifischen Serum Antikörpern. Damit stellt es die Basis für einen neuen ELISABindungsassays dar. 3\. Die immunologischen Eigenschaften dieses Antigens wurden in verschiedenen Testsystemen geprüft und damit auch charakterisiert. Die Antikörper gegen das BDV-Matrixprotein können lineare- und konformationsepitope erkennen. Die spezifische Antikörperreaktivität gegen die Epitope auf diesem rekombinanten Matrixprotein wurde mittels indirektem ELISA und Western Blot gezeigt. 4\. Antikörper gegen Matrixprotein bei Seren von verschiedenen natürlich infizierten Mammalier-Spezies zeigte eine signifikante Seroprävalenz, was dieses Protein für die seroepidemiologischen Studie der Bornaschen Krankheit in chronischem Stadium von Interesse ist. 5\. Die hohe Prävalenz von Antikörpern gegen das BDV-Matrixprotein des in humanen Seren bestätigt die Existenz des Virus beim Menschen, sowie die starke Virusaktivität und die klinische Relevanz der Bornavirus Infektion für die genannte Krankheitssymptomatik.","In the present study experiments were conducted to detect antibodies against the matrix protein in sera of humans and other species being naturally infected with the Borna Disease Virus. The following results were achieved: 1\. Sequencing of the matrix protein of different strains shows a strong phylogenetic relationship. These results confirm the stability of the virus after passages in tissue cell culture and furthermore show that the matrix protein isolated from men and animals is highly conserved. 2\. The gene encoding the BDV matrix protein was cloned in a prokaryotic expression vector. The matrix protein was over-expressed in bacteria (E. coli) and purified by Immobilized Metal Affinity Chromatography (IMAC) in high yields. The protein proved to be useful as antigen for the detection of antibodies in sera and by this constitutes the basis of an ELISA. 3\. The immunological properties of this antigen were characterized using different assays. Antibodies against the matrix protein recognize conformational as well as linear epitopes. The analysis of the antibody reactivity against these epitopes of the recombinant matrix protein was demonstrated by ELISA and Western Blot. 4\. A significant seroprevalence of antibodies against the BDV matrix protein was found in naturally infected mammalian species. This fact indicates that the matrix protein can be useful in seroepidemiological studies of infected animals during a chronic phase of the disease. 5\.
