Den Schwerpunkt stellte die Differenzierung möglicher Interaktionen zwischen VEGF und bFGF sowie potentieller Inhibitionskräfte spezifischer Rezeptorinhibitoren (SU 5416-VEGF-Rezeptorblocker/SU5402- FGF- Rezeptorblocker) dar. Als Versuchsmaterial dienten humane RPE-Zellen der ersten Zellpassage in der Zellkultur. Die Beurteilung der Wirkungsmodulation entsprechender Agenzien erfolgte zeitabhängig über die Berechnung der Zellproliferation mittels Zellzählung. Durch die Anwendung des t-Tests und der Bonferroni-Korrektur konnte signifikant belegt werden, dass beide Wachstumsfaktoren potente, zeitabhängige Stimuli der RPE-Zellproliferation sind. BFGF konnte dabei eine stärkere Wirkung zugeschrieben werden. Weiterhin lieferten die Ergebnisse Hinweise für die Existenz eines synergistisch mitogenen Effekts zwischen VEGF und bFGF. Die Applikation beider Rezeptorblocker verdeutlichte deren Inhibitionspotential, VEGF- und /oder bFGF-induzierte proliferationsfördernde Effekte signifikant zu reduzieren und einen Ansatzpunkt zur therapeutischen Intervention darzustellen. Trotz Selektivität zeigten beide Rezeptorblocker Interaktionen mit Zellen, die mit dem jeweils anderen Wachstumsfaktor inkubiert wurden. Ein Erklärungsmodell basiert auf der Vorstellung, dass die synergistische Wirkung zwischen VEGF und bFGF über einen wechselseitigen Triggereffekt realisiert wird, an dem endogen synthetisierte wie auch exogene Wachstumsfaktoren beteiligt sind. Unter Berücksichtigung bereits existierender Studien kann eine wechselseitige Upregulation über eine gesteigerte Sekretionsmenge und /oder vermehrte Expression des Wachstumsfaktors und seines Rezeptors sowie modifizierte Rezeptoraffinitäten vermutet werden. Die Ergebnisse könnten dazu beitragen, die komplexen, in vivo existierenden Interaktionen verschiedener Wachstumsfaktoren im Rahmen okulärer Pathologien besser zu verstehen.","This experimental study focused on the proliferation of RPE cells depending on the growth factors VEGF and bFGF in vitro. Key aspect was the differentiation of possible interactions between VEGF and bFGF as well as of potential inhibitory effects of specific receptor inhibitors (SU5416-VEGF-receptor blocker /SU5402-FGF-receptor blocker). Humane RPE cells of the first passage in the cell culture provided the material for the trial. The modulation of cell effects of relevant agents were evaluated time-dependently by calculation of the cell proliferation based on cell counting. By applying the t-test and Bonferroni correction it could be attested that both growth factors are capable, time-dependant stimuli of RPE cells’ proliferation. A stronger effect could be ascribed to bFGF. Further on, the results provided an indication for the existence of a synergistic mitogenic effect between VEGF and bFGF. The application of both receptor blockers showed their inhibitory potential to significantly reduce proliferating effects induced by VEGF and bFGF. This could constitute a basis for therapeutic intervention. Despite selectivity both receptor blockers interacted with cells incubated with the other growth factor. An explanatory model is based on the idea that synergistic effects between VEGF and bFGF are realized by interactive trigger effects engaging endogenously synthesized and exogenic growth factors. Considering existing studies an interactive up-regulation by an increased amount of secretion and/or increased expression of a growth factor and its receptor as well as by modified receptor affinities can be presumed.
