Die Mehrzahl der mutierten V2R weisen einzelne Aminosäureaustausche auf, die zu Fehlfaltung, Retention und schließlich zum Abbau führen. Einige V2R Mutanten werden jedoch wie der Wildtyp an der Plasmamembran exprimiert. Ziel dieser Arbeit war die Charakterisierung plasmamembranständiger, X-NDI-auslösender V2R Mutanten bezüglich Reifungsprozess, Stimulierbarkeit mit Arginin-Vasopressin (AVP) und synthetischer peptidischer Analoga; sowie die Charakterisierung des Internalisierungsverhaltens und der β-Arrestin-Rekrutierung. Durch Expression der Rezeptoren in humane embryonale Nierenzellen (HEK) und Analyse des Glykosylierungstatus konnte eine Rezeptorprozessierung und –reifung im sekretorischen Transportweg mit Insertion in die Plasmamembran für die fünf Mutanten L44F, G185C, R202C, V277A, F287L nachgewiesen werden. In Analysen der quantitativen Membranexpression des Wildtyps und der Mutanten V277A und F287L ergab sich eine vergleichbare Oberflächenexpression; bezüglich der weiteren Mutanten L44F, G185C und R202C war sie vermindert. Mittels Oberflächenbiotinylierung konnte eine deutlich ausgeprägte AVP-vermittelte Internalisierung des V2R sowie der Mutanten V277A und F287L gezeigt werden und das fluoreszenmikroskopisch beobachtete AVP-induzierte Internalisierungsverhalten bestätigen. Die Mutanten L44F, G185C und R202C zeigten fluoreszenzmikrokopisch allenfalls eine schwache Internalisierung. Auch mittels verschiedener synthetischer AVP-Analoga konnte keine Internalisierung ausgelöst werden. Bezüglich der F287L-Mutante konnte ein verändertes Internalisierungsverhalten gezeigt werden: Nach Einsatz von hohen AVP-Konzentrationen ergab sich eine deutliche Rezeptorinternalisierung. Es zeigte sich jedoch, dass β-Arrestin 2 zwar an die Plasmamembran rekrutiert wurde, aber nicht gemeinsam mit dem F287L Rezeptor internalisierte sondern an der Plasmamembran verblieb. Damit stellt die V2R Mutante F287L einen neuen Phänotyp dar, welcher offenbar AVP-induziert β-Arrestin 2 rekrutieren kann, aber β-Arrestin 2 unabhängig internalisiert. Die veränderte Interaktion der Mutante F287L mit β-Arrestin 2 weist Analogien zu dem Konzept von biased ligands auf: Biased ligands sind dadurch charakterisiert, dass die Liganden bevorzugt β-Arrestin 2 oder G-Proteine rekrutieren bzw. aktivieren. Die F287L Mutante könnte ein Prototyp des „Biased Receptors“ sein, der β-Arrestin 2 rekrutiert, aber nur schwach das G-Protein Gs und in der Folge die Adenylylcylase aktiviert.","The X-chromosomal nephrogenic diabetes insipidus (X-NDI) is a rare disease leading to symptoms concerning the electrolyte metabolism, diureses and urological complications.Most mutations lead to the misfolding of the V2R, turning it transport-defect and therefore intracellularly retained. The aim of this thesis was to characterize the disease-causing mutants L44F, G185C, R202C, V277A and F287L that are transport competent and therefore expressed at the plasma membrane. The hypothesis of this thesis was whether these mutant receptors could be stimulated by AVP or other synthetic analogues. Stimulation was observed through their internalization pattern and their ability to recruit β-Arrestin 2. Whole membrane isolates of cells expressing the V2R and the three mutant receptors L44F, G185C and R202C were digested with PNGaseF and EndoH. As expected, all the receptors were processed in the cellular secretory pathway leading to their insertion to the plasma membrane. A whole cell surface biotinylation assay of the V2R and mutants V277A and F287L demonstrated a profound internalization upon AVP stimulation. This result was consistent with the internalization after AVP stimulation observed within life cell imaging experiments with laser scanning microscopy. As expected, the other mutants L44F, G185C and R202C showed in similar experiments only a weak internalization after AVP treatment, as these mutants have been already described as AVP binding-deficient. In further life imaging experiments these mutants were stimulated with synthetic AVP-analogues. Neither of the used synthetic analogues were able to stimulate the mutant receptors L44F, G185C or R202C, but all of them stimulated as expected the V2R. With regards to the F287L mutant a different internalization pattern was observed in life cell imaging studies: Only after treatment with high concentrations of AVP, the mutant receptor showed internalization. β-Arrestin 2 was also recruited to the plasma membrane, but showed less co-localization with the receptor inside the cell and it remained longer at the plasma membrane. These experiments prove that there is further need for research in terms of receptor conformation that results in effector protein activation.
