AQP2 expression of IMCD cells kept in medium elevated to 600 mosmol/l by equimolar addition of sodium and urea (600N, control) dropped drastically when cells were instead exposed for 6 days to hypoosmolality (medium with 300 mosmol/l, 300N). No effect of medium osmolality was evident on the CMV promoter-governed AQP2 expression in WT-10 cells, suggesting that osmolality and solute composition act on AQP2 transcription rather than affecting AQP2 mRNA or protein stability. The AVP-induced translocation of AQP2 from intracellular stores to the plasma membrane appeared unaffected by osmolality and solute composition in both cell types. In IMCD cells, the phosphorylation of the transcription factor CREB increased dose-dependently with DBcAMP concentrations, whereas osmolality and solute composition did not influence CREB phosphorylation. Elevation of tonicity by sodium increased AQP2 expression in a concentration- dependent manner. Elevated urea alone was not sufficient for a robust AQP2 expression, but enhanced the effect of elevated tonicity on AQP2 expression. The effects of hypo-and hypertonicity on AQP2 expression were reversible. AQP2 expression increased slowly in response to hypertonicity, whereas hypotonicity led to a rapid decrease in AQP2 expression. In contrast, AQP2 expression increased more quickly in response to DBcAMP addition, but decreased more slowly upon DBcAMP withdrawal. A potential binding-site for the transcription factor tonicity-responsive element binding protein (TonEBP), a tonicity- responsive-element (TonE), was located within the 5´-regulatory region of the human, murine and rat AQP2 gene, with its position relative to the transcription initiation site being conserved amongst species. TonEBP expression was altered by osmolality and solute composition. Moreover, TonEBP abundance was mainly confined to the nuclei in IMCD cells kept in 600N medium, but appeared in the cytosol upon hypotonic challenge. Vice versa, hypertonicity, but not urea-derived hyperosmolalitiy, triggered a translocation of TonEBP from the cytosol to the nucleus of IMCD cells. In addition, compounds implicated in interfering with the expression of TonE- regulated genes have been tested for their influence on the hypertonicity- elicited increase in AQP2 expression. Proteasome inhibition as well as treatment with rottlerin abolished the increase in AQP2 expression expected with the onset of hypertonicity. Inhibition of p38 or ERK activation reduced the hypotonicity-elicited decrease in AQP2 expression and enhanced the promoting effect of hypertonicity. The results of this study, in conjunction with evidence accumulated in the literature, emphasize that the �classical signalling cascade� (V2R stimulation by AVP, cAMP formation, increased PKA activity, Ser-133 phosphorylation of CREB and activation of the CRE-element in the AQP2 promoter) is not the sole pathway determining AQP2 expression. This study strongly suggests by several lines of evidence that the effect of tonicity is exerted at the level of transcription via activation of the TonE/TonEBP pathway. It is proposed that the activity of the TonE/TonEBP pathway may set the margin for the effect of the classical AVP-triggered pathway on AQP2 expression in vivo||Die vorliegende Arbeit zeigt, dass in mit DBcAMP (500 µM) dauerstimulierten, primär kultivierten IMCD Zellen die Expression von AQP2 mRNA und Protein gleichartig durch die Osmolalität und die stoffliche Zusammensetzung des Kulturmediums beeinflußt wurde. Die AQP2 Expression von IMCD Zellen, kultiviert in Medium dessen Osmolalität durch equimolare Zugabe von Natriumchlorid und Harnstoff auf 600 mosmol/l erhöht wurde (600N, Kontrolle), sank nach Überführung in hypoosmolales Medium (300 mosmol/l, 300N) drastisch ab. In WT-10 Zellen (Deen et al., 1997) hatte die Medienosmolalität keinen Einfluß auf die durch einen viralen Promoter gesteuerte AQP2 Expression. Dies deuted darauf hin, dass in IMCD Zellen nicht die AQP2 mRNA -oder Proteinstabilität, sondern die AQP2 Transkription durch die Osmolalität beeinflußt wurde. Veränderungen der Medienosmolalität hatten in beiden Zelltypen keinen Einfluß auf die AVP-induzierte Translokation von AQP2 aus dem Cytosol an die Plasmamembran. In IMCD wurde die Phosporylierung des Transkriptionsfaktors CREB nicht durch die Medienosmolalität beeinflußt, nahm aber dosisabhängig mit steigenden Konzentrationen von DBcAMP zu. Eine Erhöhung der effektiven Osmolalität (Tonizität) durch membranimpermeable Osmolyte (NaCl oder Sorbitol) war eine Vorraussetzung für eine starke AQP2 Expression. Das relativ membranpermeable Osmolyt Harnstoff allein hatte keinen Effekt, verstärkte aber den Einfluß erhöhter Tonizität. Die Effekte von Hypo- und Hypertonizität waren reversibel. Hypotonizität führte zu einem raschen Absinken der AQP2 Expression, Hypertonizität löste eine langsame Zunahme der AQP2 Expression aus. Im Gegensatz dazu sank die AQP2 Expression nach DBcAMP Entzug langsamer ab und nahm bei Zugabe von DBcAMP schneller zu. In der regulatorischen Region des AQP2 Genes von Mensch, Maus und Ratte wurde jeweils eine potentielle Bindungsstelle für den Transkriptionsfaktor tonicity- responsive element binding protein (TonEBP), ein tonicity-responsive element (TonE) lokalisiert, dessen Position in Bezug zum Transkriptionsstart konserviert ist. Die Expression von TonEBP wurde durch die Osmolalität und Medienzusammensetzung beeinflußt. In Kontrollzellen löste Hypotonizität eine deutliche Verringerung von TonEBP im Kern und eine Zunahme im Cytosol aus. Hypertonizität führte zu einer Zunahme von TonEBP im Nukleus. Der durch Hypertonizität ausgelöste Anstieg der AQP2 Expression konnte durch Proteasominhibition und auch durch Rottlerin verhindert werden. Es ist bekannt, dass beide Substanzen die Expression von TonE-regulierten Genen beeinträchtigen. Bei Inhibition von p38 oder ERK wurde die durch Hypotonizität ausgelöste Reduktion der AQP2 Expression vermindert, sowie der durch Hypertonizität bedingten Anstieg der AQP2 Expression verstärkt. Die Ergebnisse der vorliegenden Studie verdeutlichen, dass die �klassische� Signalkaskade (V2R Stimulation durch AVP, Bildung von cAMP, erhöhte PKA Aktivität, Ser-133 Phosphorylierung von CREB und Aktivierung des CRE- Elements innerhalb des AQP2 Promoters) nicht allein die AQP2 Expression bestimmt. Mehrere Belege weisen darauf hin, dass die Regulation der AQP2 Expression durch Tonizität sehr wahrscheinlich über den TonE/TonEBP Signalweg vermittelt wird und die Transkription von AQP2 beeinflußt.
