Humane ET-1-DNA wird dazu mit dem chromogenen LacZ-Gen gekoppelt und in das Mäusegenom integriert. Mit Hilfe der Bluo-Gal-Färbung werden bei diesen präpro-ET-1-LacZ-transgenen Mäusen die LacZ-Markierungen in Form von blauen Präzipitaten im Nierengewebe histologisch sichtbar. Sie spiegeln die Lokalisation der ET-1-Expression im Gewebe der Mäuseniere wieder. Zusätzlich wird die NO-vermittelte in-vivo-Modulation der ET-1-Expression über die NO-Blockade durch L-NAME histologisch dargestellt und somit der Mechanismus des renalen ET-1-cGMP-NO-Regelkreises in vivo betrachtet. Der Nachweis der Aktivität des humanen präpro-ET-1-Promotors durch LacZ quantifiziert die Aktivität des ET-1-Gens effektiv in den verschiedenen Gewebestrukturen der transgenen Mäuseniere. Darüber hinaus übt der Transgenstatus keinen Einfluss auf den Phänotyp der Mäuse aus. Das Modell bietet daher die Möglichkeit, die Regulation des humanen präpro-ET-1-Promotors und somit die Regulation des ET-1-Systems auf zellulärer Ebene in vivo im Nierengewebe zu analysieren.","In this work the expression of the human prepro-ET-1-promotor shows the regulation of ET-1-expression in kidneys of transgenic mice on a cellular level in vivo. For that purpose human ET-1-DNA is attached to the chromogenic LacZ-gen and integrated into the genome of the mouse. The histologic Bluo-Gal- staining then produces blue precipitates at the location of the LacZ-activity in the renal tissue of these prepro-ET-1-LacZ-transgenic mice. The blue stains thus localise the renal ET-1-expression. In addition this work shows the NO- triggered in-vivo-modulation of ET-1-expression by suppressing NO with L-NAME, highlighting the mechanism of the renal ET-1-cGMP-NO-feedback in vivo. The detection of human prepro-ET-1-promotor activity differentiates effectively the activity of the ET-1-gene in the different kidney tissues of the transgenic mice.
